Asymbiotic germination, multiplication and development of Alatiglossum fuscopetalum (Orchidaceae) as affected by culture medium, sucrose and growth regulators

Abstract: -This investigation aimed at studying the germination and initial growth of Alatiglossum fuscopetalum (Hoehne) Baptista under in vitro conditions. The eff ects of Murashige/Skoog (MS and ½MS), Knudson (KN), and Vacin/Went (VW) culture media on the in vitro germination of A. fuscopetalum were analyzed. The infl uence of these media on the multiplication and initial growth of 90-day-old seedlings, and of diff erent sucrose, benzyladenine [BA] and indole-butyric acid [AIB] concentrations on the multiplication an… Show more

“…Four microscope slides per replication containing protocorms/seedlings were prepared and analyzed by means of a dissecting microscope. The percentages of individuals obtained for each developmental stage and for each replication were multiplied by 1, 2, 3 and 4 (weights) according to their respective stages, so that the growth index could be calculated according to Spoerl (1948), as modified by Ferreira et al (2017). The growth index of each replication was the sum of all stages of their individuals.…”

“…The most adequate conditions for orchid seed germination and, consequently, seedling growth in vitro vary according to the genus and species (Suzuki et al 2009;Abrão et al 2014;Ferreira et al 2017), which demonstrates the importance of studies regarding culture medium composition, especially its mineral nutrients, carbohydrate concentration and hormonal makeup. Such studies would allow for a better understanding of the physiological processes involved in the development of these species.…”

“…It could be noted that Brazil has the highest number of published works both on propagation and germplasm conservation, while for some countries only researches referring to propagation of native orchid species are reported and for other countries (Guyana, Paraguay, Trinidad and Tobago, Suriname, Uruguay) there were no registered works in any of the topics.
