Astroglial modulation of transient potassium current development in cultured mouse hippocampal neurons

Abstract: Hippocampal neurons exhibit three voltage-gated potassium currents, two transient currents and a delayed rectifier, that influence numerous aspects of electrogenesis including action potential duration and accommodation to sustained depolarization. These currents, termed A-, D- , and K-currents, respectively, can be distinguished based on kinetics, steady state inactivation characteristics, and sensitivity to 4- aminopyridine (see Wu and Barish, 1992b). We have compared the voltage- gated potassium currents in… Show more

“…Note that in time the specific ZK conductance increased by 64%, Wu and Barish 1992). In later whereas the specific IA conductance decreased to 15% in cells grown in KlO. on glial cells (Wu and Barish 1994). There is also evidence from rat sympathetic neurons that a fast transient current is expressed only when the neurons are cocultured with their ganglionic nonneuronal cells.…”

“…There is evidence from both in vivo and in vitro studies that potassium current expression can be regulated by extrinsic factors varying from different growth factors (Dourado and Dryer 1994;Cooper 1992, 1993;Wu and Barish 1994) to hormones (Joels and De Kloet 1992).…”

“…Developmental changes in the kinetics of voltageactivated K+ channels also occur in mouse hippocampal neurones (Wu & Barish, 1992) and amphibian spinal neurones (Barish, 1986;Harris, Henderson & Spitzer, 1989) although these later changes do not appear to be dependent upon environmental factors. However, in the case of mammalian hippocampal neurones, astroglial cells modulate the development of transient K+ currents (Wu & Barish, 1994 MMEMMaq.…”

“…Sci. USA 96 (1999) plasticity in expression of other ion channels, including K ϩ and Ca 2ϩ channels that also contribute to neuronal excitability (58,59). It is thus possible that expression of voltage-sensitive channels contributing to excitability is altered in neuronal cell types outside of the hypothalamus in response to changes in their input.…”
