Associations between genetic polymorphisms of membrane transporter genes and prognosis after chemotherapy: meta-analysis and finding from Seoul Breast Cancer Study (SEBCS)

Abstract: Membrane transporters can be major determinants of the pharmacokinetic profiles of anticancer drugs. The associations between genetic variations of ATP-binding cassette (ABC) and solute carrier (SLC) genes and cancer survival were investigated through a meta-analysis and an association study in the Seoul Breast Cancer Study (SEBCS). Including the SEBCS, the meta-analysis was conducted among 38 studies of genetic variations of transporters on various cancer survivors. The population of SEBCS consisted of 1338 b… Show more

“…For example, the human SLC12A8 gene has been identified as a psoriasis susceptibility candidate gene 33 . Most recently, it has been reported that the high expression of SLC12A8 and its genetic polymorphism are associated with better prognosis of patients with pancreatic ductal adenocarcinomas 34 and breast cancers 35 , respectively. Thus, the Slc12a8 NMN transporter could be a new, interesting target for pharmaceutical drug development.…”

“…SLC12A8 is thought to be a candidate for psoriasis susceptibility [26]. Kim et al found that SLC12A8 was correlated with a shorter overall survival of breast cancer patients, probably because the variation of SLC12A8 may be involved in the transportation of chemotherapeutic drugs, leading to a worse prognosis [19]. However, the expression of SLC12A8 in bladder cancer and its mechanism of action have not been reported.…”

“…However, the study on SLC12A8 remains limited up to now. On the basis of current researches, we found that the upregulation of SLC12A8 was related to psoriasis, psoriatic arthritis, atopic dermatitis, and breast cancer [17][18][19]. But to our knowledge, no report has been published about the biological function and the regulation of SLC12A8 expression in bladder cancer.…”

“…Therefore, ABCB1 polymorphisms do exert significant effects on breast cancer chemotherapy responses. 44 The meta-analysis results conducted by Kim et al 45 polymorphism ABCB1 in rs1045642 (C>T) was associated with poor progressionfree survival (PFS), especially in Asian patients (Hazard Ratio (HR) = 1.56, 95%, Confidence Interval (CI): 1.07-2.27). The association of rs1045642 with PFS was significant in observational studies (HR = 1.28, 95% CI: 1.05-1.56); however, this association was not significant in clinical trials (HR = 1.47, 95% CI: 0.96-2.27).…”
