The growth hormone factor-1/pituitary-specific transcription factor Pit-1 is responsible for the expression of growth hormone in mammals. Mutations in Pit-1 have been found in growth hormone disorders of mice and humans. We studied the eventual association between Pit-1 polymorphism using the HinfI enzyme and the milk yield and conformation traits of 89 Italian Holstein-Friesian bulls. A strategy employing polymerase chain reaction was used to amplify a 451-bp fragment from semen DNA. Digestion of polymerase chain reaction products with HinfI revealed two alleles: allele A was not digested (451-bp fragment), and allele B was cut at one restriction site, generating two fragments of 244 and 207 bp. Three patterns were observed; frequencies were 2.2, 31.5, and 66.3% for AA, AB, and BB, respectively. Fixed and mixed linear models were fitted on daughter yield deviations for milk yields and on deregressed proofs for conformation traits. Predictions were weighted using the inverse of the estimated variance of records. The models used contained mean and gene substitution effects for Pit-1 A allele as fixed effects and random sire effect for the mixed model. The A allele was found to be superior for milk and protein yields, inferior for fat percentage, and superior for body depth, angularity, and rear leg set, which is difficult to explain. A canonical transformation revealed that Pit-1 had three actions, one linked to milk yield traits and angularity, a second linked to body depth and rear leg set, and a third linked to lower fat yields and to higher angularity. Multitrait, across-country evaluation procedures were adapted to improve reliability of survival breeding values by combining direct information with yield and functional traits: milk persistency, somatic cell count, fertility (male and female), and calving ease (direct and maternal). A set of bulls was selected from the Austrian Simmental population based on mean original reliabilities of at least 0.50 for yield traits or of at least 0.20 for functional traits. Only breeding values above these limits were retained. The breeding values were deregressed, assuming that they were obtained by single-trait, sire-maternal grandsire models. An expectation-maximization restricted maximum likelihood algorithm based on the multitrait, across-country evaluation equations was used to compute genetic correlations among all of these traits. These equations were solved, and the reliabilities of the solutions were estimated. Mean rank correlation between direct and combined breeding values was 0.85 with values as low as 0.67 for the group of youngest bulls. Direct (original), indirect, and combined reliabilities were compared to appreciate the impact of our procedures on improvement of reliability of survival breeding values. This improvement, dependent on the level of reliability of direct and indirect information, could be up to 0.24 for animals with low direct reliability and high indirect reliability. For young bulls born in 1992 without reliable direct survival information but with already reliable information on yield and functional traits, mean reliability improvement was 0.13. For all bulls this value was still 0.06.

Associations between casein haplotypes and milk yield traits of offspring from 5 Swiss Fleckvieh AI test bulls were investigated. The analysis was performed by using a daughter design, where each daughter inherited either paternal haplotype B-A1-A-A or B-A2-A-A for alleles of alpha s1-, beta-, alpha s2- and kappa-casein genes. The substitution effects of paternal CSN2 A1 versus A2 on protein yield deviations (YDs) were significant (P < 0.05), whereas their effects on milk and fat YDs were not. The paternal substitution effects of the CSN2 A1 versus the A2 allele on protein YDs within the 5 sires did not reach the significance level. This is due to the contrary allele substitution effect of a sire compared to the other 4 sires. The effects of maternal haplotypes on milk, protein and fat YDs were not significant. However, it is noteworthy that the effects of haplotypes with a low frequency in the population deviate largely from the most frequent haplotype B-A2-A-A. The effects of beta-lactoglobulin (BLG) genotypes were significant for protein YDs but not for milk and fat YDs. The association between the paternal CSN2 A1 and A2 alleles and milk protein YDs within sires but not milk and fat YDs indicate an interaction, which might be a consequence of CSN2 heterogeneity or a closely linked gene that is contributing to the estimated effects.
