ABSTRACT. The serotoninergic system has been implicated in the etiology of schizophrenia and other behavioral disorders. Association studies have focused on the tryptophan hydroxylase 2 gene (TPH2) and the 5-hydroxytryptamine receptor 2A gene (5-HTR2A). We genotyped two single-nucleotide polymorphisms, A1438G of 5-HTR2A and intronic rs1386494 of TPH2 in the Malay population, using a sample size of 289 schizophrenic patients and 130 healthy controls. We found a significant association of A1438G of 5-HTR2A with schizophrenia in Malays. On the other hand, TPH2 polymorphism was not associated with schizophrenia. This is the first genetic association study concerning schizophrenia in the Malay population.

Ранний семейный анамнез сердечно-сосудистых за-болеваний (ССЗ) является неблагоприятным фактором для прогноза развития ишемической болезни сердца.Установлено, что при смерти одного близнеца мужского пола от ССЗ до 55 лет, риск смерти другого близнеца со-ставляет 8,1, если он монозиготный, и 3,8 для дизиготной ранний семейный анамнез сердечно-сосудистых заболеваний (ССз) является неблагоприятным фактором развития ишемической болезни сердца (иБС). установлено, что риск смерти от ССз наследуется. исследования последних лет дают убедительные доказательства, что психосоциальные аспекты личности человека играют немаловажную роль в развитии и прогрессировании болезни. на настоящий момент известно, что генотипы 5-ht2a связаны с депрессией, тревогой, агрессивно-импульсивными чертами личности, сокращением гладкомышечных сосудов, антитромботической активностью, воспалительными реакциями артерий. все вышеописанные процессы связаны с патогенезом иБС. Цель исследования -изучить связь аллелей гена 5-HT2A (а-1438G) с дебютом ишемической болезни сердца. Материал и методы. проанализировали связь психологических характеристик мужчин с учетом генотипов 5-ht2a (А-1438g) с возрастом дебюта иБС в целом и с учетом нозологической формы иБС. оценка депрессии осуществлялась психиатром по шкале Гамильтона, другие психологические характеристики оценены с помощью опросника оценки фрустрации розенцвейга, опросника тревоги Спилберга, опросника личностных черт Большая пятерка, опросника агрессии Басс-перри. результаты. участвовали 180 мужчин. Мы обнаружили, что пациенты с генотипом АА гена 5-HT2A показывают меньший уровень депрессии в сравнении с генотипом ag (p=0,021). также лица с генотипом АА имеют значительную положительную корреляцию личностной тревоги с возрастом дебюта иБС (p=0,0009, коэффициент beta=0,61), значительную положительную корреляцию личностной тревоги с возрастом дебюта иБС в виде инфаркта миокарда (иМ) (p=0,031, beta=0,52). пациенты с генотипом АА имеют возраст дебюта иБС достоверно ниже в сравнении с генотипом ag (p=0,048), с усилением достоверности различия генотипа АА от генотипов Аg и gg при сравнении среднего возраста дебюта иБС в виде иМ (p=0,021 и p=0,039 соответственно). Выводы. Мы полагаем, что связь нозологической формы, тревожно-депрессивных реакций и возраста дебюта заболевания можно проследить только у лиц с определенным генотипом. в нашем исследовании для гена рецептора серотонина типа 2a таковым является генотип (А-1438a), для которого оказался характерен ранний дебют иМ при низких баллах личностной тревоги.
