Association of tumor necrosis factor-α gene polymorphisms and coronary artery disease susceptibility: a systematic review and meta-analysis

Abstract: Background: The goal of this study was to review relevant case-control studies to determine the association of tumor necrosis factor-α (TNF-α) gene polymorphisms and coronary artery disease (CAD) susceptibility. Methods: Using appropriate keywords, we identified relevant studies using PubMed, Cochrane, Embase, CNKI, VANFUN, and VIP. Key pertinent sources in the literature were also reviewed, and all articles published through April 2019 were considered for inclusion. Based on eligible studies, we performed a m… Show more

“…Some researchers have reported that TNF-α -308G/A polymorphism is implicated in CAD development [ 37 , 38 ], whereas some others reported no evidence in this context [ 39 ]. It is suggested that the implication of the polymorphism in CAD pathogenesis differs between geographical ethnicities, and there was no significant association of TNF-α -308G/A polymorphism with the development of CAD according to a recent meta-analysis [ 26 ].…”

“…There is an SNP (Rs1800629) in the promoter region of the TNF-α (position-308G/A), where allele A at this position ( TNF-α -308A) is suggested to be associated with the higher occurrence of OSA [ 21 ] as well as with the severity of this disorder [ 18 , 22 , 23 , 24 , 25 ]. An association between TNF-α -308A allele and obesity is also reported [ 17 , 21 ], whereas there is no significant association of TNF-α-308G/A polymorphism with CAD according to a recent meta-analysis [ 26 ].…”

“…Therapeutic targeting of TNF has shown benefit in the prevention of atherosclerosis in RA (Del Porto et al 2007), which might also be the case in psoriasis (Sattar et al 2007). Interestingly, a recent meta-analysis that includes 7,697 coronary artery disease (CAD) patients and 9,655 control patients has investigated the association of TNF gene polymorphisms and CAD susceptibility (Huang et al 2020). The authors concluded that there was no association of TNF 308G/A, 857C/T, 863C/A, and 1031 T polymorphisms with CAD susceptibility.…”
