Association of the Precursor of Interleukin‐1β and Peritoneal Inflammation—Role in Pathogenesis of Endometriosis

Abstract: Abnormal proIL-1β concentration in PF and higher macrophage secretion can escalate peritoneal inflammation and endometriosis formation. The results are presented as a total IL-1β, which is a sum of proIL-1β  and IL-1β, and we believe that it reflects the actual cytokine production. The imbalance among all studied cytokines in endometriosis may be linked with an ability to transform acute inflammation to the chronic inflammation.

“…One such pathology of the peritoneal cavity is endometriosis, a condition characterised by ectopic endometrial tissue on the peritoneum that affects 10% of women of reproductive age [108]. In addition to increases in their numbers, peritoneal macrophages obtained from women with endometriosis have heightened basal production of pro-inflammatory cytokines, including TNFa, IL-1b and IL-8, and exaggerated responses to stimulation with TLR ligands, such as LPS [109,110]. Endometrial lesions are rich in immune cells and, in particular, macrophages.…”

“…Anaesthetised 6-12 week old C57BL/6J CD45.1 + CD45.2 + animals were exposed to a single dose of 9.5 Gy γ -irradiation, with all but the head and upper thorax of the animals being protected by a 2 inch lead shield. Animals were subsequently given 2-5×10 6 BM cells from sex matched or mismatched congenic CD45.2 + C57BL/6J animals by i.v. injection.…”
