Soybean cyst nematode (SCN) (Heterodera glycines Ichinohe) is one of the most economically damaging pests of soybean. The damage can only be effectively controlled through the use of resistant soybean [Glycine max (L.) Merr.] cultivars. Since QTL mapping is a highly effective approach for studying genetically complex forms of plant resistance to pests, it should be used to identify and map QTLs for resistance to SCN from various genetic backgrounds. The objective of the present study was to identify closely linked DNA markers to QTLs for resistance to SCN race 3 in the cultivar Toyomusume derived from a resistant cultivar Gedenshirazu, using the AFLP (amplified fragment length polymorphism) method combined with bulked segregant analysis (BSA). A population of 115 recombinant inbred lines (RILs) derived from a cross of Toyomusume (resistant) and Tsurukogane (susceptible) was used to map QTLs related to the resistance to SCN race 3. After genotyping of all the RILs with molecular markers including AFLP, RFLP, SSR and RAPD markers, composite interval mapping was performed for QTL analysis. In addition to known rhg1 on the linkage group (LG) G, two significant QTLs, rhg-t1 and rhg-t2, on the linkage group B1 near the AFLP markers CAT_CTC1 and GTC_ATT, respectively, were identified using two experimental assays. Epistatic analysis suggested that the combination of rhg1 and rhg-t1 could provide a higher level of resistance to SCN race 3, compared to any single QTL. The AFLP markers surrounding rhg1 and rhg-t1 could be used for markerassisted selection of SCN resistance derived from Gedenshirazu, when converted to PCR-based markers.

Soybean (Glycine max (L.) Merr.) suffers yield loss due to root infection from soil infestation by Heterodera glycine I. (soybean cyst nematode SCN) and Fusarium virguliforme (Aoki; sudden death syndrome (SDS)). The major locus for SCN and SDS resistance has previously been identified as Rhg1/Rfs2 (chr18; LG G) (site reference). The objective of this experiment was to compare the Sanger DNA sequence of a resistant cultivar ('Forrest') and two susceptible cultivars ('Williams 82' and 'Asgrow A3244'). Sequences were downloaded from GenBank for Williams 82, Phytzome for A3244 and a newly sequenced BAC-B73P06 (82,157 bp) encompassing the Rfs2/Rhg1 locus. Using the resistant cultivars, 800 single nucleotide polymorphisms (SNPs) and 57 indels were identified. In contrast, the susceptible cultivars had just 12 SNPs and no indels between them. Polymorphisms were clustered within 59 kbp, divided into three sections. There were 5 predicted recombination breakpoints. The third and fourth breakpoints were located before gene 3 and after gene 5 (Glyma18g02680; the RLK at Rhg1/Rfs2) which were therefore inferred to be derived from Peking, within the Rhg1/Rfs2 region. Comparisons of SNPs identified in Illumina sequences from 31 semi-domesticated genomes showed 80% of the total SNPs in Forrest were found among the genomes. Annotation and gene prediction showed the BAC gene prediction encoded 9-10 genes. There were 31 SNPs within exons and 137 among introns. -The objective of this work was to determine soybean resistance inheritance to Heterodera glycines Ichinohe (soybean cyst nematode -SCN) races 3 and 9, as well as to evaluate the efficiency of direct and indirect selection in a soybean population of 112 recombinant inbred lines (RIL) derived from the resistant cultivar Hartwig. The experiment was conducted in a completely randomized design, in Londrina, PR, Brazil. The estimated narrow-sense heritabilities for resistance to races 3 and 9 were 80.67 and 77.97%. The genetic correlation coefficient (r g = 0.17; p<0.01) shows that some genetic components of resistance to these two races are inherited together. The greatest genetic gain by indirect selection was obtained to race 9, selecting to race 3 due to simpler inheritance of resistance to race 9 and not because these two races share common resistance genes. The resistance of cultivar Hartwig to races 3 and 9 is determined by 4 and 2 genes, respectively. One of these genes confers resistance to both races, explaining a fraction of the significant genetic correlation found between resistance to these SCN races. The inheritance pattern described indicates that selection for resistance to SCN must be performed for each race individually.Index terms: Glycine max, Heterodera glycines, resistance, SCN, recombinant inbred lines. O experimento foi conduzido em delineamento inteiramente casualizado, em Londrina, PR, Brasil. A herdabilidade em sentido restrito para resistência às raças 3 e 9 foi de 80,67 e 77,97%. O coeficiente de correlação genética (r g = 0,17; p<0,01) demonstrou que alguns componentes genéticos para resistência às duas raças são herdados conjuntamente. O maior ganho pela seleção indireta foi obtido na raça 9, selecionando-se para a raça 3, devido à herança mais simples na raça 9, e não pelo compartilhamento de genes comuns para resistência às duas raças. A resistência da cultivar Hartwig nas raças 3 e 9 é determinada por 4 e 2 genes, respectivamente. Um desses genes confere resistência a ambas as raças, o que explica parte da correlação genética entre a resistência a estas raças de NCS. O padrão de herança descrito indica que a seleção para resistência ao NCS deve ser realizada em cada raça individualmente.
