Association of Neighborhood Disadvantage in Childhood With DNA Methylation in Young Adulthood

Abstract: IMPORTANCE DNA methylation has been proposed as an epigenetic mechanism by which the childhood neighborhood environment may have implications for the genome that compromise adult health. OBJECTIVE To ascertain whether childhood neighborhood socioeconomic disadvantage is associated with differences in DNA methylation by age 18 years.

“…Environmental conditions, both indoor and outdoor, have been linked to objective measures of the stress and relaxation response, such as heart rate variability and cortisol levels. In striking recent studies, environmental conditions have been associated with epigenetic changes in children living in disadvantaged neighborhoods ( 155 , 156 ). As most Americans currently spend over 90% of their time indoors, the critical role of building design and environmental conditions on cognitive function, sleep, stress and productivity is increasingly being realized ( 157 , 158 ).…”

“…Epigenetic mechanisms, specifically DNA methylation (DNAm), offer insights into possible molecular pathways. DNAm regulates gene expression without altering the underlying DNA sequence, is associated with health outcomes and mortality, and responsive to exogenous factors, including area-level environmental exposures [ 20 , 21 , 25 27 ]. Several measures of DNAm aging, also known as “epigenetic clocks,” have been developed using DNA microarray technology and statistical algorithms to identify alterations in DNAm that are highly correlated with chronological age [ 28 30 ].…”

“…In a study of 1,226 adults in the Multi-Ethnic Study of Atherosclerosis (MESA), the neighborhood social environment, measured as a summary score that included social cohesion, was associated with DNAm in stress-and inflammation-related genes (16). In our study, men living in neighborhoods with higher neighborhood social cohesion (15,16). Additional pathways, such as smoking status and dietary behaviors, may affect these associations.…”

“…Growing evidence suggests that accelerated biological aging at the cellular-and molecular-levels may help to explain the mechanisms linking adverse neighborhood environments to mortality (12)(13)(14)(15)(16)(17)(18)(19). Epigenetic mechanisms, particularly DNA methylation (DNAm), has offered promising insights into possible molecular pathways.…”
