Association of low‐activity ALDH2 and alcohol consumption with risk of esophageal cancer in Chinese adults: A population‐based cohort study

Abstract: Existing evidence remains inconclusive as to how the association between inactive ALDH2 and esophageal cancer (EC) depends on alcohol consumption. The study is based on the China Kadoorie Biobank cohort, with 10 years follow‐up of 0.5 million adults aged 30–79 years. ALDH2 activity was assessed by both self‐reported flushing response and Glu504Lys (rs671 G > A) polymorphism. Among both male and female participants who consumed alcohol less than weekly (n = 69,519; 211 EC cases), low active or inactive ALDH2 wa… Show more

“…ADH activity is much higher than ALDH activity, showing that cancer cells have a stronger ability to oxidize ethanol than normal tissues, but the ability to remove acetaldehyde is weak, indicating that acetaldehyde may promote cell canceration [21]. Furthermore, in East Asian populations, there is a common loss-of-function variant of the ALDH2 gene on chromosome 12 (rs671) [22,23]. This variant can lead to the loss of the catalytic activity of aldehyde dehydrogenase, which makes the body unable to eliminate acetaldehyde quickly and effectively.…”

“…Esophageal Neoplasms is one of eight most common cancers whose mortality was in top six among human cancers all over the word [30]. China has one of the highest incidence of Esophageal Neoplasms [31]. The survival rate of five-year maintained at 15% to 25% even though multimodal therapeutic means were utilized to treat Esophageal Neoplasms [32,33].…”

“…ICD code‐based diagnoses may be an incomplete representation of a patient's health status, which may also be recorded in doctor's notes and elsewhere in the EHR. In response to this problem, there exists an extensive literature on using other structured and unstructured content of the EHR to define phenotypes more accurately . Additionally, human validation can be used to evaluate phenotyping algorithms .…”
