Association of Genetic and Environmental Factors With Autism in a 5-Country Cohort

Abstract: IMPORTANCE The origins and development of autism spectrum disorder (ASD) remain unresolved. No individual-level study has provided estimates of additive genetic, maternal, and environmental effects in ASD across several countries. OBJECTIVE To estimate the additive genetic, maternal, and environmental effects in ASD.

“…It is now known that ASD is not a single disorder but one with many subtypes, and that it usually stems from multiple causes such as genetic and environmental factors. A growing body of evidence supports the role of genetics in ASD, and a heritability of about 80% has been reported (Bai et al, ). Both rare and common variants are associated with ASD, but the association of genetic risks derived from common risk variants with ASD traits in children from the general population remains unclear.…”

“…Last, most of the animal models used to study ASD are models of syndromic forms of ASD, which makes up a small proportion of ASD cases. ASD is known to have a strong genetic component (Bai et al, 2019). However that includes a variety of types of genetic variation in addition to genetically identifiable syndromes, including de novo single nucleotide and CNVs, as well as polygenic risk that results from a combination of rare and common variants (Weiner et al, 2017).…”

“…Articles we publish are also influential in news and social media, as indicated by Altmetric scores. The JAMA Psychiatry articles from 2019 with the highest Altmetric score are listed in the Table. 2- 4 One of these articles also made it onto the list of top 100 scientific articles with the highest Altmetric scores for 2019. 6 JAMA Psychiatry has a deserved reputation for publishing rigorous and sophisticated scientific research, but it also caters to a broad clinical audience.…”
