Association of creatine phosphokinase with the cytoskeleton of cultured mammalian cells.

Abstract: . Using an antibody specific for creatine phosphokinase (CPK), we have discovered an association between that enzyme and the cytoskeleton . Immunofluorescence observations show that CPK is associated with intermediate filaments in PTK cells and BALB/3T3 cells . The CPK distribution also follows intermediate filaments when cells are treated with colchicine .Intermediate (10-nm) filaments, microtubules, and actin filaments in the cytoplasm of cells contribute to a fibrous network known as the cytoskeleton. The r… Show more

“…The macrophages were enlarged and vacuolized, confirming the literature. CK is part of the stress fibers, which are located in the splenic sinus endothelium 33 . The enlarged macrophages require more space, which could be the reason for injured stress fibers that probably released CK in the surrounding area and lead to the increased plasma CK.…”

“…Overcoating of the TPV with host cell IF may also serve other functions for the parasite. Given that many ccllular organelles and macromolecules, such as heat shock proteins [ 191, protein kinase C [24] and phosphocreatine phosphate [4] are attached to the IF, envclopment of the parasitophorous compartment with IF may serve to bring nutrients and metabolites within the vicinity of the parasite. Interestingly, during adipogenesis, a n arrangement of vimentin-ER-mitochrondria occurs around the developing lipid globules [8].…”

“…Our results agree with the findings of Atkinson (1968) that the adenylate energy charge is maintained within the range of 0.8-0.9 in all viable cells. CK, particularly the MM isozyme and contractile proteins, were shown by Eckert et al (1980) to be present in some nonmuscle mammalian cell cultures. The seeming lack of correlation of mitochondrial CK and the concentration of PCr is not a t variance with the energy shuttle hypothesis.…”
