Association of castration-dependent early induction of c-myc expression with a cell proliferation of the ventral prostate gland in rat

Abstract: The protooncogene c-myc is known to be associated with both cell proliferation and apoptosis. The possible cellular affects of castration on the ventral prostate gland of rat as well as the relationship to a castration induced c-myc expression were examined. Levels of c-myc mRNA in the ventral prostate gland peaked at 6 h (early induction) and 48 h (late induction) after castration, respectively. Castrationinduced DNA fragmentation was not observed at an early induction of c-myc mRNA. DNA fragmentation appeare… Show more

“…First, c-myc mRNA levels increase following castration, as shown by multiple groups. [141][142][143] This might seem paradoxical, given the well-known role of c-myc in cell proliferation. However, it is now quite clear that c-myc can drive apoptosis under conditions where other proliferative signals (such as IGF-1) are absent or reduced.…”

“…91 It is also noteworthy that c-MYC levels are increased in the prostate after castration. 92 Furthermore, a recent study describes that overexpression of c-MYC may lead to defects in double-strand DNA break repair and induction of chromosome translocations, suggesting that c-MYC may serve as a "dominant mutator" to accelerate tumor development. 93 Microarray-based expression profiling of the c-myc transgenics has led to identification of some signature gene activities, which are shared between human and mouse prostate cancers.…”
