Association between tumor necrosis factor-α promoter −308 A/G polymorphism and susceptibility to sepsis and sepsis mortality: A systematic review and meta-analysis

Abstract: Our systematic review and meta-analysis demonstrates that the TNF2 polymorphism is associated with sepsis. However, TNF2 is not associated with sepsis mortality.

“…It is believed that these SNPs may affect gene expression so as to predispose to the development of acne vulgaris, although in some cases [8] they may offer protection from this skin disorder. These SNPs are mainly linked with predisposition to severe infections [11,12] and to the development of metabolic syndrome in women [13]. Metabolic syndrome is also a common denominator of females suffering from polycystic ovary syndrome (PCOS) which is a common endocrine disorder mediated through hyperandrogenism [14].…”

“…The best studied SNP of TNF is rs1800629 leading to one A/G transition at −308 position of gene promoter. The significance of the carriage of this SNP had been fully clarified in one meta-analysis of 25 case-association studies (28). Results showed that carriage of A alleles was associated with susceptibility to sepsis but not with mortality.…”

“…Thus, although being a carrier of the SNP TNF-308A was shown to increase the risk of sepsis in a meta-analysis that included mostly adults and older children, 44 there is so far no evidence of any association between the SNP TNF-308 G/A and increased risk of sepsis during the neonatal period.…”
