Association between the insulin-like growth factor 1 gene rs2195239 and rs2162679 polymorphisms and cancer risk: a meta-analysis

Abstract: BackgroundMany epidemiological studies have suggested that insulin-like growth factor1 (IGF1) gene single-nucleotide polymorphisms (SNPs) may be associated with cancer risk. Among several commonly studied polymorphisms in IGF1 gene, rs2195239 and rs2162679 attracted many attentions. So we perform a meta-analysis to determine potential associations between IGF1 rs2195239 and rs2162679 polymorphisms and cancer risk.MethodsWe retrieved relevant articles from the PubMed, Embase, and Web of Science databases up to … Show more

“…This may explain why IGF-1 resulted in extreme increases in axial length in chicks but has only been related to mild to moderate myopia in humans. Interestingly, rs2162679 of IGF1 had been reported to be associated with several kinds of cancer, in which the allele G could also decrease the risk of cancer onset (Xu et al, 2019). It reminded us that IGF-1 might play similar roles in myopia and cancer.…”

“…SNP rs2162679 of IGF1 has been reported to be associated with several kinds of cancer [ 40 42 ], which reminds us that IGF1 SNPs might play similar role in the onset or progesssion of myopia and cancer. In this study, our meta-analysis shows there is association between IGF1 rs2162679 and any myopia in codominant model (GA vs. AA) and dominant model (GG+GA vs. AA).…”

“…However, mutations in the exons of the IGF1 gene have been related to several SNPs that have been associated with disease states including diabetes 31,32 and cancer. [32][33][34][35][36] The expressed IGF-I protein is comprised of four domains; that is, B (49-77), the amino-terminal domain; C (78-89), A (90-110) and D (111-118), the carboxyl-terminal domain. The domains play different roles in the binding affinity of IGF-I to receptors and binding proteins.…”

“…The amino acid sequence of expressed IGF‐I is identical for all of these isoforms, as the sequence variation occurs in the extension peptide regions of the signalling (isoform 3) and propeptides (isoforms 2–4). However, mutations in the exons of the IGF1 gene have been related to several SNPs that have been associated with disease states including diabetes 31,32 and cancer 32–36 …”
