Association Between Sleep Traits and Rheumatoid Arthritis: A Mendelian Randomization Study

Abstract: Currently, the causal association between sleep disorders and rheumatoid arthritis (RA) has been poorly understood. In this two-sample Mendelian randomization (TSMR) study, we tried to explore whether sleep disorders are causally associated with RA. Seven sleep-related traits were chosen from the published Genome-Wide Association Study (GWAS): short sleep duration, frequent insomnia, any insomnia, sleep duration, getting up, morningness (early-to-bed/up habit), and snoring, 27, 53, 57, 57, 70, 274, and 42 indi… Show more

“…The odd ratio (OR) and standard error (SE) were calculated to show effect sizes. In addition, the IVW method and MR-Egger regression were used to investigate the presence of heterogeneity in the results, which was quantified using Cochran’s Q-test [ 24 ]. MR-Egger regression is also used to determine the likelihood of pleiotropy, with the intercept term indicating potential horizontal pleiotropy [ 25 ].…”

“…Various sensitivity analysis methods were used in this study. The IVW estimates approach was used to detect heterogeneity, which was measured using Cochran’s Q statistic and a p -value > 0.05, suggesting no heterogeneity [ 37 ]. The MR-Egger regression method was used to test for directional pleiotropy based on the intercept test [ 38 ].…”

“…The Weighted median method yielded consistent estimates of causal effects when approximately half of the SNPs were invalid IVs [ 30 ]. The IVW approach and MR–Egger regression were used to investigate the presence of heterogeneity in our results, which was quantified using Cochran’s Q test [ 31 ]. MR–Egger regression was also applied to determine the possibility of pleiotropy, whose intercept term represented the potential horizontal pleiotropy [ 32 ].…”
