Association between manganese superoxide dismutase (MnSOD) gene polymorphism and elderly obesity

Abstract: Evidence suggests an association between obesity and oxidative stress caused by superoxide production. Since the dismutation of superoxide is catalyzed by superoxide dismutase enzymes, we tested the association between obesity and Ala16Val manganese-dependent superoxide dismutase gene (MnSOD) polymorphism. We analyzed 815 free-living community subjects (> or =60 years old) grouped into subjects who were either obese (BMI > or = 30 kg/m(2)) or non-obese (BMI < 25 kg/m(2)). Additionally, we investigated the poss… Show more

“…A subgroup of the initial study group was asked to donate blood again for in vitro tests because a previous study performed by Montano et al (2009), has shown an association between the Ala16Val polymorphism, dietary behavior, and health conditions. The 8 mL blood samples were collected via venipuncture using tubes with heparin, which were centrifuged within 1 h of collection for 15 min at 3000 rpm.…”

“…Interestingly, in some cases, the disease risk is associated with the A allele or AA genotype -for example, in prostate cancer (Taufer et al, 2005), breast cancer (Bica et al, 2009), and other cancers. Disease risk is linked to the V allele or VV genotype in diabetes microvascular complications (Tian et al, 2011), obesity (Montano et al, 2009), hypercholesterolemia (Duarte et al, 2010), and metastatic potential of breast cancer (Bica et al, 2010).…”

“…Previously, Montano et al [14] described positive association between VV genotype and obesity and other studies also described association with hypercholes-terolemia [15], diabetes or diabetic cardiovascular diseases [16,17] diabetic nephropathy [18] and diabetic retinopathy [19], elevate plasmatic oxided-LDL levels mainly in diabetic patients [20], elevate also lipid peroxidation, protein carbonylation [10,21] and proinflammatory-cytokines (IL-1β, IL-6, TNFα and Igɣ) [22].…”

“…The elderly native group (GP group) was selected from the Southern Brazilian Aging Research Program, which investigates genetic and environmental interactions in aging and related non-transmissible diseases. Previous studies have been published and described in detail about this research project (da Cruz et al, 2003;Prado-Lima et al, 2006;Montano et al, 2009;Duarte et al, 2010). Exclusion criteria were: <60 years old and previous chronic diseases that could influence the analysis performed, such as cancer, auto-immune diseases, etc.…”
