Association between functional polymorphisms in the flanking region of miR-143/145 and risk of papillary thyroid carcinoma

Abstract: MiR-143 and miR-145 were down-regulated in papillary thyroid carcinoma (PTC) involving in cell proliferation, apoptosis, migration, invasion, and epithelial to mesenchymal transition. In this study, we aimed to investigate the association between 2 functional polymorphisms (ie, rs4705342 and rs353292) in the flanking region of miR-143/145 and risk of PTC. A case-control study including 316 PTC patients and 347 controls was performed. The rs4705342 and rs353292 were genotyped by using the TaqMan alle… Show more

“…The TT/CT genotypes of rs353292 were found to be associated with a higher risk of chronic kidney disease and colorectal cancer (15,16). However, there was no significant correlation between the rs353292 polymorphism and susceptibility to papillary thyroid carcinoma and conotruncal heart defects (25,26). In the present study, our results also demonstrated that the genotype frequencies of rs353292 were not significantly different between T2DM patients and controls.…”

“…Systematic search; duplicate removal; and screening of titles, abstracts, and full text yielded eight eligible articles [ 41 , 43 , 44 , 45 , 46 , 47 , 48 , 49 ]. The expression of miR-145 was significantly reduced in thyroid cancer compared with normal cells [ 41 ].…”

“…Overexpression of miR-145 may reverse the EMT phenotype of thyroid cancer cells; relative levels of miR-145 in thyroid cancer tissues were significantly lower than in the corresponding normal tissues [ 46 ]. Moreover, Wang et al, testing genetic variants flanking the miR-145 gene, found significant association between rs4705342*T single nucleotide polymorphism and a higher risk of papillary thyroid carcinoma [ 48 ].…”

“…The association between rs4705342 and cancer risk has been examined extensively, but the results were controversial. The C allele of rs4705342 was found to play a protective role against the occurrence of papillary thyroid carcinoma (18). However, Kotarac et al (19) reported that the rs4705342 C variant was associated with an increased risk of prostate cancer.…”

“…To date, several polymorphisms have been identified in the miR-143/145 cluster. Among these polymorphisms, two widely studied polymorphisms (rs4705342 and rs353292) have been reported to be correlated with the risk of tumor development and cancer progression (17)(18)(19)(20). In this study, we evaluated the effects of the two genetic variants on the risk of EOC and the prognosis of EOC patients from North China.…”
