Association between cyclooxygenase-2 (COX-2) 8473 T > C polymorphism and cancer risk: a meta-analysis and trial sequential analysis

Abstract: BackgroundNumerous studies have investigated the relationship between COX-2 8473 T > C polymorphism and cancer susceptibility, however, the results remain controversial. Therefore, we carried out the present meta-analysis to obtain a more accurate assessment of this potential association.MethodsIn this meta-analysis, 79 case-control studies were included with a total of 38,634 cases and 55,206 controls. We searched all relevant articles published in PubMed, EMBASE, OVID, Web of Science, CNKI and Wanfang Data, … Show more

“…The meta-OR for the 8473 T>C polymorphism in the proinflammatory cyclooxygenase-2 (COX2) gene was 0.65 under the recessive model, based on three studies of NPC (two in Caucasians and one in Asians; ref. 189). Across four NPC case-control studies in Asian populations, for the -94 ATTG insertion/deletion polymorphism in the NFkB subunit 1 gene (NFKB1), the meta-OR was 0.78 under the recessive model (190).…”

“…They are essential in developing malignant and benign conditions [ 19 ]. More specifically, cyclooxygenase enzymes (COX), MMPs, and prostaglandins can inhibit tumor suppressor genes through DNA methylation and post-translational modification, which potentially results in cancer development [ 63 ].…”

“…COX-2 is an inducible enzyme whose expression is associated with increased inflammation and various pathological processes [100]. COX-2 is induced by numerous proinflammatory stimuli including inflammatory cytokines and monocyte cells themselves [101]. COX-2 catalyzes the synthesis of prostanoids (PGE2, TXA2) from arachidonic acid and serves as the main source of these molecules during inflammation [102].…”
