Association Between Cortical Superficial Siderosis and Dementia in Patients With Cognitive Impairment: A Meta-Analysis

Abstract: Background: It remains unclear whether cortical superficial siderosis (cSS) is associated with dementia and its subtypes. We thus performed a meta-analysis to evaluate the relationship between dementia and cSS.Methods: We searched EMBASE, PubMed, and Web of Science for relevant studies assessing risk of dementia and prevalence of cSS in patients with cognitive impairment. Fixed-effects and random-effects models were performed. Results: Seven eligible studies including 3,218 patients with definite cognitive impa… Show more

“…The same findings were observed in a meta-analysis of subjects with cognitive impairment, in that there is an association between the presence of cSS and Alzheimer’s disease. 39 Currently, there is limited evidence from a mechanistic viewpoint to explain the relationship between cSS and cerebral tau accumulation. This observation could be suggestive of a possible relationship between leptomeningeal Aβ vasculopathy and cerebral tau pathologies, as cSS has been associated with a higher severity of CAA in the leptomeningeal vessels, but not in the cortical vessels.…”

“…Although ARIA-E has been observed mostly during clinical trials of Aβ-lowering therapeutics [13 , 17 , [24] , [25] , [26] , [27] , [28] , [29] ], it may also develop spontaneously in AD patients, likely indicating the inflammatory phase of cerebral amyloid angiopathy [30] . Similarly, microhemorrhages and superficial siderosis spontaneously develop in cerebral amyloid angiopathy patients, regardless of anti-Aβ therapy [31 , 32] .…”
