Association Between a Novel Variant of the Human Type 2 Deiodinase Gene Thr92Ala and Insulin Resistance

Abstract: Thyroid hormone action is an important determinant of energy and glucose metabolism. T4 metabolism is regulated by the deiodinases of which type 2 is expressed in humans in skeletal muscle and brown adipose tissue, where its transcription is stimulated by the ␤-3 adrenergic pathway. We performed molecular scanning of the human type 2 deiodinase (DIO2) gene and evaluated a novel variant for associations with obesity and insulin resistance, assessing both the main effect and interaction with the Trp64Arg ␤-3-adr… Show more

“…Focus was then turned to the KPF induction of Dio2. D2 is an important component of the thermogenic gene expression profile in stimulated BAT and probably in HSM (3,(21)(22)(23). First, we verified that Dio2 induction by KPF resulted in a 8.5-to 50-fold increase in D2 activity in more than one cell line, including HSM myoblasts, MSTO-211H cells, and RMS-13 cells (Fig.…”

“…The common Thr92Ala D2 polymorphism (7) has been associated with traits of impaired thyroid hormone action at various end-organ targets (7-10), the hypothalamic-pituitary level (11), and in circulating levels of thyroid hormones (12). Nonetheless, results are conflicting (13)(14)(15)(16) and the data refer to steady-state serum thyroid hormone levels.…”

“…Data reporting an association between this change and the risk of hypertension (120,121) or insulin resistance/diabetes are conflicting (58,(122)(123)(124). In a recent large case-controlled study of 1057 diabetics and 516 controls, the frequency of Thr92Ala was higher in the diabetic group.…”
