Association Analyses with Carcass Traits in the Porcine KIAA1717 and HUMMLC2B Genes

et al.

Abstract: By screening a subtracted cDNA library constructed with mRNA obtained from the longissimus dorsi muscles of F1 hybrids Landrace×Yorkshire and their Yorkshire female parents, we isolated two partial sequences coding for the H3-K4-specific methyltransferase (KIAA1717) and skeletal muscle myosin regulatory light chain (HUMMLC2B) genes. In the present work we investigated two SNPs, one (C1354T) at the 3' untranslated region (UTR) of KIAA1717 and one (A345G) at the SINE (PRE-1) element of HUMMLC2B, in a resource po… Show more

“…In some cases both genes and the underlying causal mutation have been identified by the candidate gene approach (Harlizius and van der Lende, 2001;Li et al, 2002;Xu et al, 2005). Animal breeders have started applying markerassisted selection to improve the quality and performance of livestock (van der Steen et al, 2005) and genetic polymorphisms (marker loci) significantly associated with important traits have become very useful tools.…”

“…However, the marbling scores, which reflect the levels of Several quantitative trait loci (QTL) for meat quality traits including backfat and total body fat (Rohrer, 2000;Edwards et al, 2008) and physiological traits in blood such as alkaline phosphatase activity (Yoo et al, 2012), CD4positive leukocyte number (Wattrang et al, 2005), and bilirubin levels (Reiner et al, 2008) (www.animalgenome.org/ cgi-bin/QTLdb) are mapped at the region on Sus scrofa chromosome 8 (SSC8) where SETD7 is located. Reports of an F 2 population of an intercross between Large White and Chinese Meishan pigs described a significant difference in the levels of backfat thickness (Xu et al, 2007), marbling score, and intramuscular fat percentage (Xu et al, 2005), according to the SETD7 genotypes. Lee et al (2011) also reported a significant difference in various traits, including backfat thickness, in a comparison of each genotype.…”

“…A set of primers designed by Xu et al (2005) was used for SETD7 gene amplification and genotyping. PCR was performed using 25 ul of reaction mixture including 100 ng of DNA, 1.0 nmole of each primer, and 2.5 U of i-Taq DNA polymerase (Intron Biotechnology, Korea).…”
