Assignment of the major histocompatibility complex to the p1.4→q1.2 region of chromosome 7 in the pig (<i>Sus scrofa domestica</i> L.) by in situ hybridization

Abstract: The pig major histocompatibility complex (SLA) was mapped by in situ hybridization. The probe used was a human cDNA containing most of the coding sequence for HLA-B7. The SLA complex was localized to the region p1.4→q1.2 on chromosome 7.

“…Molecular analysis of the tumour necrosis factor gene region in swine has revealed sequences characteristic of the MHC found in other species (Kuhnert et al 1991). This finding, along with linkage studies (Vaiman et al 1990) and analysis of a panel of sorted chromosomes (Chardon et al 1990), proves that TNFA is on porcine chromosome 7 and also suggest that TNFA and TNFB map to the porcine MHC region (SLA), found to be located within porcine 7~12-12 (Rabin et al 1985;Echard et al 1986). Here we present data allowing the regional assignment of TNFA and TNFB on porcine chromosome 7.…”

“…Subsequent analysis using the same probe (Echard et al 1986) or a full-length porcine class I gene probe (PDIA; Rabin et al 1985;Singer et al 1988) reported the SLA to lie near the centromere in the region 7p1.4-q1.2 and 7p1.2-q1.2, respectively. The low resolution of these assignments could result from a dispersed distribution of SLA genes that crosshybridize with the selected probe; however, a limited physical map of the SLA produced via pulsed field gel electrophoresis (PFGE) suggests the complex is -2 Mb in width (Xu et al 1992).…”

“…Previous physical assignments of polymorphic microsatellite loci on SSC7 were limited to three markers in a 22-cM region in the vicinity of the centromere (Rabin et al 1985;Echard et al 1986;Poulsen et al 1991;Solinas et al 1992;Ellegren et al 1994a;Yerle et al 1995). Two RFLP markers have been physically assigned but only expand the physical map to 25 cM (Geffrotin et al 1984;Rabin et al 1985;Echard et al 1986;Yerle et al 1995).…”

“…By in situ hybridization the loci for the SLA could be assigned to chromosome no. 7 (G EFFROTIN et al , 1984 ;R ABIN et al , 1985 ;E CHARD et al, 1986). Therefore the linkage group C-J (M UIR & R ASMUSEN , 1974) is likely to be located on the same chromosome, since a close linkage has been found between the loci for the blood group J and the SLA (H RUBAN et al, 1976).…”
