Assessment of tolerance to multistresses and in vitro cell adhesion in genetically modified Lactobacillus plantarum 590

Abstract: Lactobacillus plantarum (Lp) is a lactic acid bacterium that has many excellent traits that meet the needs of industrial production. Genetically modified (GM) Lp590 was obtained from Lp that was modified by the insertion of the gene nisI, which can confer resistance to nisin and play a role as a bio-preservative. Here, explorations were made to assess the safety of GM Lp590 and establish an in vitro evaluation model. The ability of Lp590 to tolerate both environmental stresses (such as temperatures ranging fro… Show more

“…A high degree of flocculation was observed for this bacterium in all tested media. A similar phenomenon was shown for other LAB species . Possibly, the formation of flocs can reduce the surface exposed to ethanol and thus help the bacteria to survive the stressful conditions of the ethanol process.…”

“…Standard curves were established as described by Rinttil et al (16). Primer sequences targeting 16SrDNA of different groups or genera of bacteria have been designed and optimized (14,18) and are listed in Table 1. Briefly, specific DNA from the two standard bacterial strains was extracted and diluted to 10-fold serial concentrations ranging from 10 10 to 10 2 DNA copies/lL according to the average genome size of the specific bacteria and ignoring differences in rrn copy numbers.…”

“…Genetically modified Lactobacillus plantarum 590 (GM Lp 590) derived from electroporation of pLEB590 as a food-grade vector (2) into L. plantarum showed that nisI function as an immunity protein in Lactobacilli, enabling nisin resistant (14,15). Microorganism intake can more directly effect on GI microbiota, their metabolites (9, 10), gut permeability, and mucosal immunity (3,16).…”

“…In addition, genetic manipulation can influence enzyme activity during EPS biosynthesis, leading to the overexpression of the UDP-glucophosphorylases and uridyl-transferases . Genetically modified (GM) Lactiplantibacillus plantarum 590 (Lp 590), obtained from Lp that is modified by inserting the gene nisI , indicates excellent antioxidant capacity and enhances resistance to oxidative stress . Heterologous expression of small heat shock protein shsp gene from Streptococcus thermophilus and RecO gene from Lactobacillus casei enhances the survival of strains under oxidative stress .…”
