BackgroundHedychium coronarium Koen. (Zingiberaceae) is traditionally used as medicine in countries such as India, China, and Vietnam for treatment of various ailments including cancer. However, in spite of its implied significance in cancer treatment regimes, there are no reports so far involving the anticancerous attributes of H, coronarium ethanol extract (HCEE) on cancer cells and a more comprehensive study on its mechanism is still lacking.Materials and methodsThe cytotoxicity of HCEE was evaluated by MTT and clonogenic survival assay. Annexin V/propidium iodide (PI), Hoechst 33342 staining, and TUNEL assay were performed to detect apoptosis. Cell cycle analysis was performed using PI staining. JC-1 and 2′,7′-dichlorodihydrofluorescein diacetate assay were used to check the levels of MMP and ROS, respectively. Western blot analysis was carried out to measure the expression levels of proteins. Migration and invasion activity were assessed by wound healing and Transwell membrane assay, respectively.ResultsAntiproliferative effect of HCEE was investigated in various cancerous and normal cell lines. Among these, HCEE significantly inhibited the survival of HeLa cells without affecting the viability of normal human umbilical vein endothelial cells. Annexin V/PI, Hoechst staining, and TUNEL assay showed HCEE induced apoptosis in HeLa cells in a dose-dependent manner. HCEE promoted cell cycle arrest at G1 phase in HeLa cells by upregulating the levels of p53 and p21 and downregulating the levels of cyclin D1, CDK-4, and CDK-6. Moreover, HCEE treatment upregulated the expression of Bax and downregulated the expression of Bcl-2. Additionally, HCEE activated the caspase cascade by increasing the activities of caspase-9, caspase-8, and caspase-3. The expression levels of Fas ligand and Fas were also upregulated. Further, HCEE inhibited the migratory potential of HeLa cells by downregulating MMP-2 and MMP-9 expression levels.ConclusionOur results indicate H. coronarium exerts antiproliferative and apoptotic effects against HeLa cells, and therefore may be used for treatment against cervical cancer.

Medicinal plants have been used in all cultures as a source of medicine since ages. When parts of plants such as rhizomes, leaves or barks and other natural materials are used as drugs to treat illnesses, they are called crude drugs and the study of crude drugs is called pharmacognosy. Proximate analysis in plants gives valuable information and help to assess the quality of a crude drug. Beneficial effects of crude drugs are believed to be attributed to plant phytochemicals. Hedychium coronarium J. Koenig, a medicinal and aromatic plant of high value is an endangered and red-listed plant. The rhizome of this plant is used for the treatment of various diseases. Crude extracts prepared from the rhizomes show antibacterial and antifungal properties. Essential oils obtained from Hedychium coronarium are found to be rich in terpenes and used for bactericidal, fungicidal, medicinal and cosmetic applications. The present study assesses the quality of crude drugs prepared from this highly medicinal plant. Proximate analysis and phytochemical screening of rhizomes of Hedychium coronarium, which is frequently consumed as food and as medicine were carried out. Ash content was found to be low) when compared to the moisture content. The water extractive value was more than alcohol extractive. Preliminary tests carried out on phytochemicals revealed the presence of terpenoids and oils. High Resolution Liquid Chromatography Mass Spectrophotometry was carried to ascertain the different components of essential oils and revealed the presence of eucalyptol (1,8-cineole), caryophyllene oxide, camphor, linoleic acid, ricinin, phloroglucinol, 6-gingerol, carvone and arjungenin.

Zingiberaceae is the largest family of the order Zingiberales containing more than 1600 species and 200 genera among which Hedychium is the medium sized ornamental genus containing near about 40-50 speciesmostly distributed throughout the tropics and sub tropics of the world. Being aromatic and rich in diverse bioactive compounds plants of this family are economically, ethnomedicinally and pharmaceutically very important. Apart from their traditional use as spices, food, ethnomedicine, dyes, perfume etc., they have got a huge future potential in new drug development due to their antimicrobial, anti-inflammatory, anti-diabetic, anti-cancer and antioxidant characteristics. In the present study, the ethnomedicinal uses of Hedychium coronarium has been documented. In addition to this the antimicrobial properties of this plant have been examined by disc diffusion method. It has been concluded that H. coronarium exhibited remarkable anti-bacterial property against the gram-positive bacteria Staphylococcus aureus than gram-negative bacteria Escherichia coli. The GCMS analysis reveals the presence of 18 compounds which are responsible for the medicinal properties of this plant.
