Assessment of the genotoxicity of heavy metals inPhaseolus vulgaris L. as a model plant system by Random Amplified Polymorphic DNA (RAPD) analysis

Abstract: Impact assessments of environmental pollutants are important in eco-genotoxicology. A random amplified polymorphic DNA (RAPD) technique was used to detect genotoxicity-induced DNA damage in Phaseolus vulgaris L. from heavy metals at two different concentrations. The results from six 10-base pair (bp) random RAPD primers with 60-70% GC content used, showed a total of 295 RAPD fragments of 700-4000 bp in molecular size in the seedlings of untreated and treated samples, of which only 163 fragments were polymorphi… Show more

“…Cd toxicity also affects the plasma membrane of plants, which could be attributed to electrolyte leakage [58], and membrane proteins like H-ATPase inhibition [42]. It has been known to affect the DNA repair system [59]; thus, the stability of the genomic template was distinctly reduced in Phaseolus vulgaris [60] and peas [61] in response to the direct application of Cd.…”

“…У проростках пшениці Triticum aestivum L. зміну RAPD-спектрів відмічали при вирощуванні в присутності 5 мг/мл (~25 мкM) хлориду кадмію впродовж 9 діб [31]. Мутагенну дію кадмію на Phaseolus vulgaris L. спостерігали при культивуванні проростків у присутності Cd(NO 3 ) 2 в концентрації 150 мг/мл (0,61 мМ) впродовж 7 діб [32].…”

“…Mazhoudi et al (1997), Mocquot et al (1996) with control which can be attributed to the fact that much metal was not accumulated by plants and remained in rhizospheric zone so toxic effects didn't reach the thresholds to make any apparent DNA damage. The number of band appeared or disappeared in samples exposed to metals suggests different metals probably cause alterations at genomic level and therefore samples exposed to heavy metal stress would act as a marker for assessment of environmental exposition on metals Gjorgieva et al (2013). Plant mechanism effecting the root uptake and shoot transport of Cd can also affect the expression of Cd toxicity in plants Cakmak, (2000); Dunbar et al (2003).…”
