Assessment of Salt Tolerance in Transgenic Potato Carrying AtNHX1 Gene

Abstract: Salt tolerance can be assessed with plant morphological and physiological traits. This study evaluated the salt tolerance of eight transgenic potato (Solanum tuberosum L.) lines using an integrated evaluation index where plant height, leaf area expansion rate, survival rate, and tuber yield were integrated. We hypothesized that (i) the integration of several plant growth traits produced a comprehensive evaluation of salt tolerance, and (ii) the level of salt tolerance in potato was also related to their physio… Show more

“…The PCA method has been used to analyze huge amounts of data and excavate valuable information, and the analytic result provided help for screening tolerant germplasm resources in crop improving programs (Fleta-Soriano and Munne-Bosch, 2016;Mahajan et al, 2013;Noreen and Ashraf, 2009;Ullah et al, 2017;Wang et al, 2013). A comprehensive system that included morphological, physiological, and biochemical components should be established for the precise assessment of stress tolerance in crops.…”

“…Detailed methodology was described previously (Wang et al, 2013). In brief, an evaluation index was developed using the following three steps:

Data standardization, where a subordinator function was formed to standardize the measured raw data of each parameter as follows: eqnarrayright center leftμfalse(Xijfalse)=false(Xij-Xminfalse)false(Xmax-Xminfalse) eqnarrayright center leftμfalse(Xijfalse)=false(Xmax-XijXmax-Xminfalse), where, Xij is the value of the j th variable (daily gain of plant height, leaf area expansion rate, stem thickness expansion rate, plant tissue water contents, tuber yield per plant, and rate of plant survival to drought treatment.)

“…To quantify the degree of drought tolerance, we employed an integrated evaluation index where a cluster analysis was combined with standard deviation coefficient allocation weighted method. Detailed methodology was described previously (Wang et al, 2013 ). “…Although some factors such as CAT activity in pea varieties, root proline content in Plantago maritima, leaf MDA level and SOD activity in soybean, and shoot dry weight in wheat were used to distinguish the salt tolerance (Noreen and Ashraf 2009;Zhang et al 2011;Sharbatkhari et al 2013;Sleimi et al 2015), the limitations of using a single evaluation indicator had been discussed (Mwadzingeni et al 2016). PCA was a comprehensive evaluation method that used widely to assess different resistant germplasm resources (Wang et al 2013). In this study, the salt tolerance of four kiwifruit genotypes were assessed by PCA, and Acva-3 had the strongest tolerance to salt stress.…”
