Assessment of Protein C Anticoagulant Pathway by Thrombin Generation Assay in the Presence of Endothelial Cells

Abstract: 3394 The generation of thrombin is the pivotal event in the process of blood coagulation. In vivo, thrombin generation is regulated by cooperation between the vascular endothelium and the pro- and anti-coagulant systems in blood, such as the thrombin/endothelial cell-dependent activation of the protein C anticoagulant pathway that ultimately leads to inactivation of factors Va and VIIIa. In vitro, thrombin generation is assessed most commonly in platelet-poor plasma by a fluorogen… Show more

“…The kinetics of thrombin and subsequent APC formation are comparable to the results obtained by continuous thrombin generation measurement in plasma in the presence of purified TM, with the exception that peak levels were significantly lower. 27 Both thrombin and APC formation declined after 60 minutes, indicating that thrombin and APC are cleared in the plasma matrix by endogenous inhibitors and that thrombin-TM complexes are internalized by ECs. 28…”

“…We performed additional experiments to further investigate an autoimmune mechanism interfering with the endothelial protein C-protein S-thrombomodulin (PC-PS-TM) system. To this end, we used a modified fluorogenic thrombin generation assay, which assesses thrombin generation in the presence of EA.hy926 cells, as previously described [4]. In this assay, thrombin generation is highly regulated by the EA.hy926 PC-PS-TM system.…”
