Powdery mildew caused by the ascomycete Phyllactinia guttata (syn. P. corylea) is a major foliar disease worldwide of the unique mulberry (Morus spp.) for silkworm feed. Genetic resistance to powdery mildew, the most sustainable and economical strategy for disease control, is still elusive for tropical mulberry. About 147 germplasm sources, representing 18 countries of origin, were screened for resistance to P. guttata in six seasonal fields and greenhouse trials after exposure to natural and artificial inoculum, respectively. In the field, the level of plant responsiveness to disease was assessed from 30 to 62 days after pruning in each season as variations in the disease severity index (DSI), disease incidence (DI%) and area under the disease progress curve (AUDPC). These measures differed significantly among the germplasm. Of 147 germplasm sources, *6.8% had useful resistance (two high and nine moderately resistant) to the powdery mildew pathogen on the basis of DSI. The AUDPC values were 13.5-fold higher in the most susceptible accession-(Philippines) than the least responsive (Vietnam-2). The results of DSI were strongly correlated with the obtained DI values (r = 0.92; P \ 0.01) and AU-DPC (r = 0.89; P \ 0.01). Moreover, field screening results were highly correlated (R 2 = 0.839) with values from the greenhouse evaluation using artificial inoculum. However, the DSI values in field and greenhouse screenings for three sources (Non-nayapati, Nao-khurkul and Tista Valley) varied significantly. A relatively low disease reaction of 09 resources (Vietnam-2, Ankara and 07 others) using different assessment scales after natural and artificial inoculation prove, for the first time, that they have potential in breeding for resistance in tropical mulberry to powdery mildew.

Black spot disease, incited by the fungus Diplocarpon rosae Wolf, is the most important disease of roses (Rosa hybrida L.) in the outdoor landscape. Though partial resistance exists in cultivated germplasm, the genetic basis of this trait has not yet been elucidated. Six diploid and six tetraploid rose cultivars were crossed in two factorial combining ability arrays. Whole plant and detached leaf inoculation methods were used to assess partial resistance under two different disease pressures using a characterized single-spore isolate. Parents from both arrays had significant general combining ability effects across multiple inoculation methods and environments. Specific combining ability was not significant for either array. Parent per se performance was highly correlated with progeny performance on a family mean basis. High positive correlations among whole plant and detached leaf inoculation methods indicate that detached leaf assays can substitute for whole plant assays. Based on these results, a breeding strategy including parental selection and early, among-family selection is proposed. To our knowledge, this is the first investigation of combining ability for disease resistance in rose.

Control of fungal diseases is a major constraint of cut-rose cultivation in greenhouses and in transportation around the world. Therefore, development of resistant cultivars is a promising way to reduce the use of chemicals required for controlling the diseases. Genetic analyses and breeding for resistance, however, are hampered by the high degree of heterozygosity and the polyploid nature of cultivated roses. Nucleotide-binding site (NBS) profiling of Van der Linden et al. (2004) was used as a tool enabling a more directed way of studying the genetics of resistance to pathogens responsible for diseases such as powdery mildew. NBS profiling is a multiplex screening technique, producing amplified resistance gene (R-gene) and resistance gene analogue (RGA) fragments by using degenerated primers based on the conserved motifs present in the NBS domain of resistance genes. Since RGAs are abundantly distributed and highly polymorphic within the plant genome, NBS profiling generates multiple markers of putative resistance genes. Twelve NBS degenerated primer/ restriction enzyme combinations were used to genotype the whole rose tetraploid K5 population (Yan, 2005) and its parents. To generate RGA profiles, the restriction enzymes: AluI, HaeIII, MseI, and RsaI were used in combination with degenerated primers NBS1, NBS3, and NBS5a6. The profiles were dominantly scored resulting in 106 polymorphic RGA markers which segregated in a 1:1 or 3:1 ratio. Uni-and biparental simplex markers will be mapped on the two available AFLP/SSR K5 maps (Yan, 2005) with Joinmap 4.0. The resulting parental tetraploid maps will be used to dissect the genetic variation for resistance to powdery mildew resistance. Additional Rosaceae SSRs mentioned in the literature are currently tested on the K5 population to obtain allelic bridges between the tetraploid and diploid genetic maps in rose and related species in order to align them. These bridges will improve cross-ploidy comparisons in roses in order to strengthen cut rose breeding.
