Assessment of MDA efficiency for genotyping using cloned embryo biopsies

Abstract: The possibility to genotype embryos prior to implantation would have advantages for increasing the speed of selection of cattle. Reliable genotyping requires more DNA than can be obtained from biopsies of embryos, if they are to remain viable. Multiple displacement amplification (MDA) is a whole genome amplification technique used to increase the amount of DNA from biopsies for analysis. Reduced genome coverage resulting in Allele Drop Out (ADO) at heterozygous loci or missing genotypes are drawbacks of MDA. T… Show more

“…The number of IVP embryos transferred has quadrupled over the past decade and now constitutes 40% of total embryos transferred in cattle. One driver for the current surge in interest in IVP embryos is the prospect of genomic selection of Day 7 embryos (Lauri et al 2013). This can significantly reduce the breeding interval (and so increase response to selection) by virtue of the fact that selection occurs at Day 7 of gestation and not at term.…”

“…The coupling of whole genome amplification and DNA sequencing have allowed multiple groups to study the genetics of single cells, but not without significant amplification biases (9)(10)(11). Moreover, single-cell exome sequencing confirmed the clonal heterogeneity of a solid tumor identifying key mutations across much of the genome (12).…”
