BACKGROUND: Resistance to insecticides has been related to application history, genetic factors of the pest and the dynamic within the treated area. The aim of this study was to assess the geographic variation in azinphos‐methyl response and the role of esterase and cytochrome P450 monooxygenase enzymes in codling moth populations collected within different areas of the Río Negro and Neuquén Valley, Argentina.RESULTS: Diapausing field‐collected populations showed resistance ratios at the LC50 that were 0.7–8.7 times higher than that of the susceptible strain. Mean esterase (EST) and cytochrome P450 monooxygenase activities (expressed as α‐N min−1 mg−1 prot−1 and pg 7‐OHC insect−1 min−1 respectively) were significantly correlated with LD50 values from the field‐collected populations. In addition, azinphos‐methyl response was associated with the geographic area where the insect population was collected: populations from isolated and more recent productive areas presented significantly lower resistance ratios in comparison with populations from older and more intensive productive areas.CONCLUSION: The populations assayed presented different resistance levels to azinphos‐methyl. The response was highly correlated with the orchard's geographic location. EST and ECOD activities were involved in azinphos‐methyl response in the given region. Functional characterization of a novel λ‐cyhalothrin metabolizing glutathione S‐transferase, CpGSTe3, from the codling moth Cydia pomonella

BACKGROUND Recent work has shown that two codling moth (Cydia pomonella) glutathione S‐transferase genes (GSTs), CpGSTd1 and CpGSTd3, can metabolize λ‐cyhalothrin, one of the recommended insecticides for C. pomonella control worldwide. However, systematical characterization of delta and epsilon GSTs, especially their potential contributions in the metabolism of λ‐cyhalothrin, is currently still lacking in C. pomonella. RESULTS In this study, a total of nine cDNA sequences were identified in C. pomonella, including four in the delta and five in the epsilon subclasses. RT‐qPCR showed that seven GSTs were ubiquitously expressed at all developmental stages, and CpGSTe2, CpGSTe3, and CpGSTe4 were mainly expressed in larvae. The mRNA levels of CpGSTd2, CpGSTd4, and CpGSTe5 were significantly higher in male than in female adults. Tissue‐specific expression analysis revealed that the CpGSTe2, CpGSTe3, and CpGSTe4 were highly expressed in the midgut while CpGSTd2 and CpGSTd4 were predominantly expressed in the Malpighian tubules. The transcripts of these GSTs (except CpGSTe1) were co‐expressed following exposure to LD10 of λ‐cyhalothrin for 3 h. Recombinant CpGSTd4, CpGSTe2, and CpGSTe3 proteins expressed in Escherichia coli displayed glutathione‐conjugating activity toward 1‐chloro‐2,4‐dinitrobenzene. In addition, λ‐cyhalothrin could inhibit the activity of recombinant CpGSTd4, CpGSTe2, and CpGSTe3 enzymes, but only recombinant CpGSTe3 showed λ‐cyhalothrin metabolic capacity, with 21.88 ± 1.09% of parental compound being depleted within 1 h. CONCLUSION These data show that CpGSTe3 is a third GST gene, encoding an enzyme that metabolizes λ‐cyhalothrin in C. pomonella. © 2019 Society of Chemical Industry

This paper describes for the first time the capacity of allyl cinnamate to elicit antennal response and to modify behavior from C. pomonella and L. botrana adults. Improved understanding of the role of this chemical compound may ultimately be incorporated into Tortricidae integrated pest management programmes (IPM). Material and Methods InsectsThe experiments were conducted with a C. pomonella laboratory strain originated from a population collected in an unsprayed apple orchard in Lleida (north-east Spain), and with a L. botrana laboratory strain established in our laboratory from a mass-reared strain from INRA Bordeaux (France). Both strains were reared at the Crop Protection Laboratory of the UdL-IRTA Centre for Research and Development (Lleida, Spain) at 23 ± 2°C, with a photoperiod of 16:8 (L:D) and on agar-based semi-synthetic diet [33].Cydia pomonella individuals were sexed as last instar larvae, whereas L. botrana individuals were sexed as pupae. The sexed individuals were kept separately by sex until adult emergence in the same conditions as AbstractElectroantenographical (EAG) response to allyl cinnamate were assessed on virgin and mated Cydia pomonella and Lobesia botrana adults to determine whether this compound could be used within integrated management programs (IMP). Adult behavioral reaction was later assessed in a wind tunnel, with and without the main compound of the corresponding female sex pheromone.Allyl cinnamate elicited antennae responses of C. pomonella and L. botrana, both males and females. Allyl cinnamate EAG response was as high as pheromone response, and it was not reduced after mating.In wind tunnel assays, allyl ester itself was not attractive to C. pomonella males, but its presence did not interfere with the pheromonal action when the number of contacts was compared. For females, a higher proportion of codling moths moved towards the source when allyl cinnamate was in the wind-tunnel plume. No differences were recorded depending on the mating status of codling moth adults. The same trend was observed in L. botrana males and females.Results suggest that allyl cinnamate acts as a female behavioral modifier, but more assays are required to determine its role in insect communication in field conditions before inclusion in integrated pest management.
