Bioluminescent Vibrio fischeri Assays in the Assessment of Seasonal and Spatial Patterns in Toxicity of Contaminated River Sediments

Several bacteria-based assays, notably Vibrio fischeri luminescence assays, are often used as environmental monitoring tool for toxicity in sediments that may serve as both sinks and secondary source of contamination in aquatic ecosystems. In this study, we used 30-s kinetic bioassays based on V. fischeri to evaluate the toxicity associated to sediments from five localities with different contamination inputs (Morava River and its tributary Drevnice River in the south-eastern part of the Czech Republic). Toxicity assessed as half maximal inhibitory concentration (IC50) over the course of a year-long sampling was compared in bottom sediments and freshly trapped particulate material. Standard approach based on testing of aqueous elutriates was compared with toxicity of whole sediments (contact suspension toxicity). Bottom sediments showed lower toxicity compared to freshly trapped suspended materials in all cases. On the other hand, standardized elutriates induced generally weaker effects than suspended sediments likely due to losses during the extraction process. Toxicity generally increased during winter reaching maximum peaks in early spring months in all five sites. Total organic carbon (TOC) was found to be highly correlated with toxic effects. Toxicity from sites with direct industrial and agricultural water inputs also correlated with concentrations of metals, polycyclic aromatic hydrocarbons (PAHs), and polychlorinated biphenyls (PCBs). Single time point sampling followed by the extraction and testing of elutriates, do not truly reflect the spatial and temporal variability in natural sediments and may lead to underestimation of ecotoxic risks.

Summary Sediment microbial fuel cells (SMFCs) are different from microbial fuel cells because they are completely anoxic and lack a membrane. SMFCs are a novel technology for the simultaneous production of renewable energy and bioremediation of heavy metals. Recently, SMFCs have attracted the attention of many researchers because of their moderate functioning parameters and ability to use a range of biodegradable substrates like glucose, glutamic acid, river water, cysteine, acetate, and starch. The inocula used in SMFCs include river sediment, marine sediment, and wastewater. For power generation, many exoelectrogens in SMFCs have the ability to transfer electrons from electrodes by using natural electron shuttles. Exoelectrogens use four primary pathways to transfer electrons to the electrodes, including short‐range electron transfer through redox‐active proteins, soluble electron shuttling molecules, long‐range electron transport by conductive pili, and direct interspecies electron transfer. The most dominant mechanism is long‐range electron transfer via conductive pili because pili have metal‐like conductivity. The powering by microbes is an emerging technique for the remediation of heavy metals from sediments. The pathways for transferring electrons in electrotrophs operate in the opposite direction from those in exoelectrogens. To further upgrade SMFC technology, this review targets the prototype, operating factors, working mechanisms, applications, and future perspectives of SMFCs. Copyright © 2017 John Wiley & Sons, Ltd.

A wide range of biological activities of aminophosphonates predisposes them to find applications as anticancer, antiviral, antimicrobial, antifungal, or herbicidal agents. Despite a number of positive aspects of the use of aminophosphonates, their applications may cause a risk to the environment, which is well exemplified by the case of glyphosate. Therefore, scientists see a pressing need to rate ecotoxicity of aminophosphonates. Nowadays, it is recommended to use comprehensive tools to carry out appropriate and effective risk assessments of toxic substances. For these purposes, tests based on the acute toxicity of the luminescent bacteria Aliivibrio fischeri, as well as the measurement of sub-chronic toxicity of the crustacean Heterocypris incongruens seem to be the most convenient. A series of five diphenyl N-arylamino(pyrrole-2-yl)methylphosphonates was synthesized and preliminary evaluation of their ecotoxicological properties was performed. In order to carry out such investigations, we applied the two biotests mentioned above. Results showed that the N-(4-nitrophenyl) derivative was the most toxic for bacteria in comparison to other tested compounds. As for crustaceans, N-phenyl and N-naphthyl derivatives were found to be the most harmful, simultaneously being relatively harmless for bacteria. Such a phenomenon are discussed in correlation with the literature, while its reason is discussed with respect to the aspect of structure of the tested compounds.
