Assessment of genetic stability on in vitro and ex vitro plants of Ficus carica var. black jack using ISSR and DAMD markers

Abstract: In vitro propagation has been signi cant in producing a large number of genetically stable regenerated plants. Regenerated Ficus carica var. Black Jack plantlets were established using woody plant medium (WPM) supplemented with 20 µM 6-Benzylaminopurine (BAP) and 8 µM Indole-3-acetic acid (IAA) under different light treatments such as normal uorescent white light (60 µmol.m − 2 .s − 1 ), and four different LED spectra, white (400-700nm), blue (440nm), red (660nm) and blue + red (440nm + 660nm). Genetic stabili… Show more

“…Our results are consistent with a recent report 31 on genetic stability evaluation of in vitro raised Ficus carica var. Black Jack plantlets using woody plant medium containing 20 μM BAP + 8 μM IAA under different light treatments (normal fluorescent white light and four different LED spectra) that showed significant (97.87%) similarity using ISSR markers.…”

“…Molecular markers have been frequently utilized for capturing the genetic variations induced during in vitro propagation or conservation of plants in form of discrete banding patterns. PCR-based single primer amplification reaction (SPAR) methods including random amplified polymorphic DNA (RAPD) and inter simple sequence repeats (ISSR) have been regularly used for genetic fidelity assessment of in vitro propagates 27 31 . The SPAR methods need no prior sequence information, are simple, economic, rapid and require small quantities of DNA 27 .…”

“…The biotechnological interventions in vitro may cause somatic variation [35,36], suggesting the need for genetic stability assessment [19,37]. The genetic stability is usually estimated via morphological identification and molecular marker assessments [19,[37][38][39]. In order to confirm the genetic stability of the present shipping system, we further compared the morphology and reproduction of recovered plantlets, which germinated in vacuum-packed microplates supplemented with 100 mg/L CCC.…”

“…SCoT markers have been reported for assessing the genetic fidelity of several plant species such as Cleome gynandra [97], Bauhinia racemosa [81], and Rauwolfia tetraphylla [38]. Similarly, the use of RAPD and ISSR markers for assessing the genetic fidelity of Ficus carica micropropagated plants was reported by several studies [34,40,87,98]. However, this is the first report using RAPD, ISSR, and SCoT primers for the assessment of the genetic fidelity of micropropagated F. palmata plants.…”
