Welding, a fabrication process that joins metals or thermoplastics by causing coalescence, is indispensable in modern society and ubiquitous in industry. Welding generates fumes that contain several metals and gases that comprise fine and ultrafine particles with the potential for adverse effects. Although health risks of welders have been evaluated in different populations, occupational exposure to welding fumes is still considered to be an important health problem, especially in developing countries. The aim of this study was to investigate the effects of welding fume exposure on important oxidative stress parameters such as superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), total glutathione (GSH), glutathione peroxidase (GPx), malondialdehyde (MDA), and 8-hydroxy-2′-deoxyguanosine (8-OHdG) in Turkish welders ( n = 48). The influence of confounding factors such as age, smoking habits, alcohol consumption, and duration of exposure on the studied parameters was also analyzed. In our study, significant decreases in the levels of GSH and activities of CAT, SOD, and GPx and significant increases of MDA, 8-OHdG levels and GR activity were found in the workers compared to the controls. There was a negative correlation between GSH levels and alcohol usage. Also, older workers (≥35 years) had significantly higher GR levels than younger workers. But smoking and alcohol usage, duration of exposure, and utilization of protective measures had no significant effect on the studied parameters in the workers. These results indicate that occupational exposure to welding fumes appears to induce oxidative stress and oxidative DNA damage.

This study aimed to assess the association of exposure to particle-bound (PM2.5) polycyclic aromatic hydrocarbons (PAHs) with potential genotoxicity and cancer risk among children living near the petrochemical industry and comparative populations in Malaysia. PM2.5 samples were collected using a low-volume sampler for 24 h at three primary schools located within 5 km of the industrial area and three comparative schools more than 20 km away from any industrial activity. A gas chromatography–mass spectrometer was used to determine the analysis of 16 United States Environmental Protection Agency (USEPA) priority PAHs. A total of 205 children were randomly selected to assess the DNA damage in buccal cells, employing the comet assay. Total PAHs measured in exposed and comparative schools varied, respectively, from 61.60 to 64.64 ng m−3 and from 5.93 to 35.06 ng m−3. The PAH emission in exposed schools was contributed mainly by traffic and industrial emissions, dependent on the source apportionment. The 95th percentiles of the incremental lifetime cancer risk estimated using Monte Carlo simulation revealed that the inhalation risk for the exposed children and comparative populations was 2.22 × 10−6 and 2.95 × 10−7, respectively. The degree of DNA injury was substantially more severe among the exposed children relative to the comparative community. This study reveals that higher exposure to PAHs increases the risk of genotoxic effects and cancer among children.

In general, an occupation could have hazzard and risk of exposure to genotoxic chemical compounds. These compounds could lead to micronucleus formation on buccal mucosa. This study was aimed to obtain the impact of chemical exposure on micronucleus formation in buccal mucosa at various occupations. This was a literature review study using databases of PubMed, ScienceDirect, and Google Scholar based on the keywords. The criteria of literatures were articles published in 2016-2021 using Indonesian or English language. The results showed that group compounds of polycyclic aromatic hydrocarbons (PAH), metals, carbamate, and organophospate, also compunds of silica, bezene, toluene, xylene (BTX), sevoflurane, desflurane, isoflurane, nitrous oxide, gemcitabin, and 5-fluoro uracil were genotoxic chemical compunds and could cause micronucleus formation in buccal mucosa of mechanics, grillers, miners, e-waste recyclers, construction workers, road markers, car painters, gasoline station workers, farmers, and healthcare workers. Genotoxic chemical compounds could be found excessively in occupational environment. These compounds could damage cells’ DNA and caused micronucleus formation on buccal mucosa of workers. Hasil penelitian mendapatkan senyawa kelompok polycyclic aromatic hydrocarbon (PAH), logam, carbamate, dan organophospate, serta senyawa silica, benzene, toluene, xylene (BTX), sevoflurane, desflurane, isoflurane, nitrous oksida, gemcitabin, dan 5-fluoro uracil merupakan senyawa kimia yang bersifat genotoksik dan dapat memicu pembentukan mikronukleus di mukosa bukal mekanik, pemanggang daging, penambang, pendaur ulang sampah elektronik, pekerja konstruksi, pekerja marka jalan, pengecat mobil, petugas SPBU, petani dan tenaga kesehatan. Senyawa kimia yang bersifat genotoksik banyak ditemukan di lingkungan pekerjaan. Senyawa tersebut dapat merusak DNA sel sehingga terbentuk mikronukleus di mukosa bukal pekerja.
