Assessment of DNA Damage by Comet Assay in Lymphocytes of Workers Occupationally Exposed to Petroleum fumes

Abstract: The comet assay is a sensitive, reliable and rapid method for DNA double & single strand break, alkali-labile sites and delayed repair site detection in individual cells. In recent years, this method has been widely used for studies of DNA repair, genetic toxicology, environmental biomonitoring, and apoptosis. However, this technique serves as an important tool for detection of DNA damage in living organisms and is increasingly being used in genetic testing of industrial chemicals, agrochemicals & pharmaceutic… Show more

“…Everatt, et al , also found no significant correlation between duration of employment and level of DNA damage by the comet assay. 8 Singh, et al , in a study on workers exposed to petroleum fumes, 15 and Costa, et al , in another study on a group of formaldehyde-exposed workers, 16 found similar results. The effect of age, as a comorbidity in comet assay results, is questionable.…”

“…Such DNA damage can be also enhanced by confounding factors like length of exposure, smoking and drinking of coffee, tea and alcohol. This result is in good agreement with earlier reports (Zhao et al, 1998;Heuser et al, 2002;Zhu et at., 2003;Müller et al, 2004;Knudsen et al 2005;Singh et al, 2010;Prasad et al, 2013). Fuel emissions have carcinogenic properties since they contain dangerous substances that are able to react with and change the genetic material, of the affected organisms.…”

“…Comet assay is a simple, rapid and reliable technique for the detection and quantification of DNA damage. The assay determines the extent of DNA damage in individual cells and this can be assessed from the length of DNA migration [15].Comet assay was performed to analyse the extent of DNA damage caused by zerumbone and cisplatinon cancer cells and normal human lymphocytes. The tail length, % tail DNA and olive moment are considered as DNA damage parameters [20]. [21,22].…”
