Assessing Mitochondrial Redox Status by Flow Cytometric Methods: Vascular Response to Fluid Shear Stress

Abstract: Mitochondria are an important source of superoxide production contributing to physiological and pathological responses, including vascular oxidative stress that is relevant to cardiovascular diseases. Vascular oxidative stress is intimately linked with pro‐inflammatory states and atherosclerosis. Oxidized low‐density lipoprotein (OxLDL) modulates intracellular redox status and induces apoptosis in endothelial cells. Hemodynamic, specifically, fluid shear stress imparts both biomechanical and metabolic effects … Show more

“…Mitochondrial superoxide formation was determined by the uptake of MitoSoxÔ Red reagent (Invitrogen) in live cells, as previously described [36]. Briefly, cells were plated at a density of 10,000 cells/cm 2 in six-well plates, with or without EET-A, 10 mM, overnight and then incubated with Hoechst nuclear stain (5 mg/mL) for 30 min in complete medium.…”

“…The fold change in mean florescence intensity both of SLE and control samples were measured. 19 ÁÉm Mitochondrial oxidant generation is regulated by Ácm, which is a marker for ROS-mediated apoptosis. The JC-1 MitoScreen assay (BD Biosciences) was used to assess Ácm.…”
