Assembly and disassembly of the Golgi complex: two processes arranged in a cis-trans direction.

Abstract: . We have studied the disassembly and assembly of two morphologically and functionally distinct parts of the Golgi complex, the cis/middle and trans cisterna/Trans network compartments . For this purpose we have followed the redistribution of three cis/middle-(GMP-,, GMP-2 , MG 160) and two trans-(GMP,-, and GMP,-2) Golgi membrane proteins during and after treatment of normal rat kidney (NRK) cells with brefeldin A (BFA). BFA induced complete disassembly of the cis/middle-and trans-Golgi complex and translocat… Show more

“…However, the close juxtaposition of several types of organelles in the perinuclear region of PrP224AlaMYC cells makes a precise localisation difficult. Therefore, we added brefeldin A (BFA) to chronically prioninfected PrP-224AlaMYC cells to break up the Golgi apparatus into dispersed vesicles termed 'ministacks' (Alcalde et al, 1992). BFA treatment enabled us to visualise the degree of colocalisation of PrP Sc with GM130 with a higher spatial resolution.…”

“…Since the effect of BFA on protein synthesis is minimal, secretory proteins tend to accumulate within the mixed ER/Golgi compartment (12,17). Remarkably, the effects of BFA have been shown to be completely reversible upon removal of the drug, with reassembly of the Golgi apparatus and resumed secretion (18,19).…”

“…Studies have, therefore, been restricted to analysis of Golgi reassembly after treatment with drugs such as Brefeldin A (11)(12)(13)(14), although it is not clear to what extent this is a suitable model for biogenesis.…”
