Assembly and disassembly of Aspergillus fumigatus conidial rodlets

Abstract: The rodlet structure present on the Aspergillus fumigatus conidial surface hides conidia from immune recognition. In spite of the essential biological role of the rodlets, the molecular basis for their self-assembly and disaggregation is not known. Analysis of the soluble forms of conidia-extracted and recombinant RodA by NMR spectroscopy has indicated the importance of disulfide bonds and identified two dynamic regions as likely candidates for conformational change and intermolecular in… Show more

“…This rodlet layer is exclusively composed of hydrophobins encoded by the RODA gene. This protein is characterized by the presence of eight cysteine residues capable of forming four disulfide bridges and organizing the protein crystal in an amyloid configuration that waterproofs the conidial cell wall (471). How the individual proteins aggregate to form a rodlet layer remains unknown.…”

“…Three sites, including K50, K55, and K126, were detected with biotinylation marks. All three lysine residues are localized in the relatively hydrophilic α-helical regions of the protein ( 33 ), suggesting that only the surface-exposed lysine residues are likely to be biotinylated.…”

“…fumigatus strain used in this study was CEA17_Δ akuB KU80 . The RodAp-cysteine residue point-mutant strains and the RODA deletion mutant were generated as described earlier [ 9 , 10 , 11 ]. All the strains were maintained on 2% malt–agar slants at ambient temperature.…”

“…Differentiation of DCs was confirmed by the expression of CD1a and the lack of surface expression of CD14 and CD16 [ 20 ]. DCs were stimulated with conidia or cell wall polysaccharides as described earlier [ 1 , 9 , 21 ]. β-(1,3)-Glucan and α-(1,3)-glucan were used at equal absolute concentrations (1 μg); they varied in their chain size, thus ruling out the possibility of using them at equimolar concentrations.…”
