Assay of the Heat-Labile Enterotoxin of Escherichia Coli in Infant Rabbits

Abstract: S T R A I N S o f Escherichiu coli enteropathogenic for swine have been shown to secrete enterotoxins in vitro (Smith and Halls, 19673;Gyles and Barnum, 1969). These enterotoxins cause accumulation of fluid when they are introduced into ligated intestinal loops prepared in rabbits or pigs (Smith and Halls, 1967~). The strains of E. coli involved are believed to produce two different types of enterotoxin, namely a heat-labile non-dialysable immunogenic toxin (LT) and a heat-stable dialysable toxin (ST).The use… Show more

“…Since H ϩ ,K ϩ -ATPase expression and gastric acid secretion are upregulated during fetal development and neonatal maturation (4,36), we examined the maturation of CA IX expression in the proximal gastrointestinal tract. In two separate rabbit litters, tissue was isolated from two to three rabbits at each time point, and tissue from one of the rabbit pairs was used for examination of CA IX mRNA abundance by quantitative real-time RT-PCR, whereas the other was used for analysis of protein expression by Western blotting.…”

“…Enterotoxin identification methods can be performed using animal models (rabbit ileal loops method) [15] and cell lines, referred to as the golden standard in LT toxin expression detection. The method using cell lines detects the cytopathic effect caused by the enterotoxin on the CHO and Y1 cell line (abnormalities of cell shape) [6,16,17].…”

“…In a recent paper by Burgess et al (1979) describing the assay of heat-labile enterotoxin of Escherichia coli in infant rabbits, the authors discussed an attempt to increase yields of enterotoxin by the method of lincomycin induction, as originally reported from this laboratory (Levner, Wiener and Rubin, 1977). I feel obliged to take issue with certain conclusions arrived at by these authors which might unjustifiably discourage other workers from using this convenient technique.…”

“…H. LEVNER Department of Research and Development, Wyeth Laboratories, Philadelphia, Pa 19 101, USA Dr Levner is correct to point out that we compared toxin yields from lincomycin-resistant mutants of strain P307 grown in the presence of lincomycin, with the yield from strain P307 grown conventionally (Burgess et al 1979).…”
