Assay of Growth Hormone Levels in Human Plasma using Commercial Kits: Analysis of Some Factors Influencing the Results

Abstract: ABSTRACT. The reliability of growth hormone (GH) assays, performed in 39 different laboratories, using five different immunoassay kits was evaluated. It was found that the variability in GH levels measured by different commercial assay kits may be due to human factors, as well as differences between the kits. The influence of the variation in amount of circulating GH, during provocative testing and spontaneous secretory episodes, on the results of GH assay was also evaluated. It was found that large molecular… Show more

“…Our results support the idea that the GH heterogeneity contributes to the discrepant results obtained when serum GH levels are measured by different assays (44)(45)(46). Isoforms and fragments lack part of the GH molecule, resulting in variable crossreactivity to monoclonal or polyclonal antibodies with distinct epitope specificity (47).…”

“…In comparative studies, it has been demonstrated that great differences exist between laboratories using the same GH kit [17], These discrepancies have partly been attributed to local factors. In some countries, great effort is made to reveal these differences [18].…”

“…As it is well known that GH concentrations vary according to the assay method [7][8][9][10][11], we have standardized the GH values measured with these commercial kits. The study described here was performed in 1996.…”
