Assay of Gamma-Glutamyl Transpeptidase Activity in Amniotic Fluid Offers a Possible Prenatal Diagnosis of Biliary Atresia in the Rat Model

Abstract: Muller et al. analysed gamma‐glutamyl transpeptidase (GGTP) activity in the amniotic fluid of more than 2000 pregnant women for a prenatal diagnosis. They reported that at 18–19 weeks' gestation, two fetuses associated with lower amniotic fluid GGTP levels were diagnosed after birth as having biliary atresia (BA). If low GGTP values correlate closely with BA, chemical assay of amniotic fluid GGTP could possibly be used in the prenatal diagnosis of BA. A fetal model of cholestasis in the rat was created by the … Show more

“…Because extrahepatic biliary atresia is present in all reported cases, we suggest that amniotic fluid ␥-glutamyltranspeptidase biochemical testing could be used in association with ultrasound studies for prenatal diagnosis in high risk families [Muller et al, 1991;Hosoda et al, 1997].…”

“…Drug administration during pregnancy can induce experimental BA in some animal models [ 58 , 59 ]. A number of postnatal maneuvers (intrabiliary injection of sclerosants and superglue) in a variety of animal models can evoke early neonatal cholestasis [ 60 67 ].…”

“…Hosada et al administered intraperitoneal phalloidin, an actin-binding toxin derived from the Death Cap mushroom ( Amanita phalloides ) to Wistar rats on gestational day 15. After birth, the pups presented with cholestasis and an increased volume of actin filaments around bile ducts, but without typical BA features [74]. In another study, the anti-helminthic drug, 1,4-phenylenediisothiocyanate, was given orally to pregnant, newborn and adult Wistar rats.…”
