Assay of D-dimer based on immunofiltration and staining with gold colloids

Abstract: In this immunofiltration assay of D-dimer in plasma samples, the antigens are captured by a monoclonal antibody on a porous membrane, and labeled with the same antibody conjugated to gold colloids. The assay time is < 2 min, and a color of intensity proportional to the concentration of D-dimer is left on the membrane. The reference range (mean +/- 2 SD) was 0.336 +/- 0.133 mg/L (n = 69). Linearity was found up to 10 mg/L. Comparison with ELISA results (x) for 198 patients' samples demonstrated a linear … Show more

“…Gold colloids produce an intense red staining that is proportional to the amount of D-dimer antigen present. 64 The intensity of the color is then compared with a reference color chart with five different color zones corresponding to D-dimer concentrations of 500, 1000, 2000, 4000 and 8000 ng/mL. 38 SEMINARS IN RESPIRATORY AND CRITICAL CARE MEDICINE-VOL.…”

“…21 There may be interference from rheumatoid factor, cellular material, heparin, or large lipid particles; however, the assay was not affected by bilirubin, hemoglobin, fibrinogen, soluble fibrin, fibrinogen degradation products, or repeated freeze-thawing. 64 The first clinical application of the Nycocard assay in a series of 92 inpatients and outpatients with suspected DVT found it to be 100% sensitive with an NPV of 100%. 21 Several studies done subsequently yielded sensitivities of 85 to 100% and NPV ranging from 0 to 100%.…”

“…In the 1970s a number of epitopes of the late products of thrombolysis were defined in particular fragments DD (D‐Dimer) and DE (fragment E) 29,30,34 . At the same time monoclonal antibodies were becoming widely available allowing these neo‐antigens to be targeted 30,35 .…”
