Assay of adenosine in human adipose tissue

Abstract: A method is described to assay the adenosine content of human adipose tissue. Tissue pieces of 10\p=n-\20 mg were frozen within 1 sec of excision by a conchotome. The pieces were pooled and ground in liquid nitrogen, divided into samples weighing 50\p=n-\100 mg each and placed in perchloric acid. Neutralized defatted extracts were treated by Dowex AG-1 to remove nucleotides and lyophilized. The adenosine content was measured by RIA using antiserum raised against laevulinic acid ( 02' , 3' \ x=r eq-\ adenosine-… Show more

“…As far as we know, this study is the first to describe high plasma adenosinelevels inoverweight/obesewithnormal pregnancyduring their third trimester of gestation. These results agree with previous reports in obese adults showing high adenosine levels in adipose tissue homogenates [17]. We also found high plasma adenosine levels in obese children [18].…”

“…In this regard, no reports concerning plasma adenosine levels in overweight/obese pregnant women have been published. But, high concentrations of this molecule were found in the homogenates of adipose tissue obtained from obese adults [17] or in plasma from obese children [18].…”

“…Using this method, amounts of adenosine as low as 1 pmol per sample could be determined. The intra-assay variability was 11.7% (N = 9) and the inter-assay variability 14.9% (N = 5) (Ranta et al 1985).…”

“…The tissue pieces, weighing 10-20 mg, were frozen in liquid nitrogen within one second of excision by a conchotome. Adenosine was assayed from pooled samples as described in detail previously (Ranta et al 1985). Briefly, the pieces were first ground in liquid nitrogen and placed in perchloric acid.…”

“…In other experiments, 10-50 mg pieces were removed rapidly with tongs precooled in liquid N2. The pieces were ground in liquid N2 and their adenosine contents were assayed by radioimmunoassay as described elsewhere (Ranta et al, 1985).…”
