Aspects of bile secretion in the rabbit.

Abstract

SUMMARY1. Bile secretion was studied in anaesthetized rabbits from whom hepatic bile was collected by cannulation of the common bile duct.2. The flow and composition of bile formed by rabbits anaesthetized with urethane differed significantly from that formed by rabbits anaesthetized with pentobarbitone sodium.3. The i.P. injection of a hypertonic solution of sucrose (3 M) decreased bile flow and produced changes in the ionic composition of bile and of plasma.4. The infusion of sodium taurodeoxycholate (1.5-20 ,umole/min I.v.) gave higher rates of bile flow than did equimolar infusions of sodium taurocholate, and unlike taurocholate, increased the bicarbonate concentration of bile.5. Acetazolamide (10-100 mg/kg) increased the concentration of bicarbonate both in bile and in plasma, and had little effect on bile flow.6. The infusion of bromsulphthalein (5 mg/kg I.v.) decreased the excretion of bicarbonate into bile, and was associated with the formation of a hypotonic bile.7. The implications of these results in relation to the mechanisms of bile secretion are discussed.

Effect of pentobarbital or urethane on bile secretion and chemical composition of blood in the rabbit

“…The bile flow values were significantly higher after pentobarbital than after urethane. Our data for the former agree with those found in the literature (Erlinger et al, 1970;Rutishauser & Stone, 1975;Gonzalez et al, 1983;Esteller et al, 1984) with one exception (Bruss, Cornelius & Himes, 1983) and are also similar to those reported for other anaesthetics such as propanidid (Shaw & Heath, 1975). With regard to urethane, although data appearing in the literature are not uniform, they are consistently lower than those found by us for stimulation of the spleen or the administration of a-adrenergic agents activates~-phosphorylase kinase and hepatic glycogenolysis (Proost, Carton & De Wulf, 1979).…”

“…The potassium plasma concentration was lower for both agents than has been reported pentobarbital (Klaassen, 1974;Rutishauser & Stone, 1975;Esteller, Lopez & Murillo, 1977). The bile-to-plasma [14C]erythritol ratios were similar to those reported by Klaassen (1974) with urethane and by Erlinger et al (1970) with pentobarbital and point to a slight ductular reabsorption.…”

“…Bile bicarbonate concentrations proved to be considerably higher with both agents compared with plasma levels; this phenomenon has not been observed in other animal species such as the rat, although data have been presented to this effect for the dog (Klaassen, 1974). It has been observed that biliary bicarbonate excretion is not depressed by acetazolamide in the rabbit and that the hepatic carbonic anhydrase of this species is refractory to this drug (Rutishauser & Stone, 1975). It appears that the mechanism involved in bicarbonate secretion in the rabbit exhibits certain peculiar characteristics.…”
