The Amazon basin houses a particular group of freshwater organisms, whose study tells the geological history of the region, how biological diversity was shaped, how it is maintained, and what it hides. The fish of the Amazon is represented by more than 3,000 known species distributed from the basal groups up to the more specialized ones. This species richness hides a diversity of adaptations that are dispersed at all levels of the biological organization. In this summary of the conference delivered at the Brazil-France Bilateral Symposium, held in Manaus in 2018, we describe two aspects of the hidden world of adaptive diversity: adaptations to changes in dissolved oxygen and the abilities of the Rio Negro fish to face acidic and ion-poor waters of their habitats. Also, we present the vulnerability of Amazonian fish to ongoing climate changes. Then, very briefly, we call attention to the many hidden biological processes that allow these fish species to survive their environments, much of which is unknown. Indeed, however, they are intricately related to men, either by responding to environmental disturbances that we have caused, or by containing information that contributes to improving the quality of the environment in which we live.

Blood-O2 affinities (P50 ) were measured over a physiologically relevant pH range at 31 (highest temperature average of Rio Negro over the last 8 years), 33 and 35° C for 10 species of the Rio Negro, aiming to test the acute effects of temperature foreseen by the IPCC (Intergovernmental Panel on Climate Change) for coming years. The animals were collected during an expedition to the Anavilhanas Islands of the Rio Negro, 110 km upstream from Manaus (2° 23' 41″ S; 60° 55' 14″ W). Hoplias malabaricus showed higher blood-O2 sensitivity to pH changes (Bohr effect, Φ = Δlog10 P50 ΔpH(-1) ) at both 31° C (Φ = -0·44) and 35° C (Φ = -0·26) compared to Osteoglossum bicirrhosum (Φ = -0·54 at 31° C and Φ = -0·58 at 35° C), but lower P50 under most conditions, and a greater sensitivity of P50 to temperature. Two out of the 10 analysed species had significant increases of P50 (lower blood-O2 affinity) at the highest temperature throughout the pH range tested. For all other species, a minor increase of P50 over the assay-tested temperatures was observed, although all presented a normal Bohr effect. Overall, a diversity of intensities of pH and temperature effects on blood-O2 affinities was observed, which seems to be connected to the biological characteristics of the analysed species. Thermal disturbances in their habitats, likely to occur due to the global warming, would impair blood-O2 binding and unloading in some of the analysed fish species. Haemoglobins of five tilapiine species of the genera Oreochromis and Sarotherodon were investigated. By gel filtration chromatography a molecular weight of 67-69 kDa was determined for the tetrameric molecules which remained stable between pH 5.0 and pH 9.1. When subjected to sodium dodecyl sulphate-Ureapolyacrylamide gel electrophoresis (PAGE), haemoglobins of all species each were split into monomers of three different molecular weights ranging between 16.3 kDA and 17.6 kDa. Subsequently, isoelectric focusing separated haemolysates into about 23 differently charged tetrameric haemoglobins that were arranged in species-specific patterns. This diversity was shown to result from the occurrence of different types of globin chains. By acidic urea PAGE a total of seven major alpha-globins and five major beta-globins were detected and species-characteristic chain variants were identified. To determine the globin chain composition of particular haemoglobin tetramers, 26 bands were isolated by isoelectric focusing and analysed by acidic urea PAGE. Tetramers consisted of doublets of identical alpha- and identical beta-chains (alpha 2 beta 2, symmetric tetramers), or combinations of three (alpha 2 beta beta *; alpha alpha * beta 2) or four (alpha alpha * beta beta *) distinct chains (asymmetric tetramers). Finally, globin chains of Oreochromis niloticus were subjected to partial N-terminal amino acid sequencing. Differences in the composition of the three major beta-chains could be shown, whereas the alpha-chains were N-terminally blocked.
