Asiatic acid enhances survival of human AC16 cardiomyocytes under hypoxia by upregulating miR‐1290

Abstract: Asiatic acid (AA) could attenuate ischemia/reperfusion induced myocardial apoptosis through upregulating the Akt/GSK-3β/HIF-1α pathway. HIF-3α is a negative regulator of HIF-1α, whose mRNA is a potential target of miR-1290. AA could upregulate miR-1290 in non-small-cell lung cancer A549 cells. This work aimed to investigate whether AA could inhibit hypoxia induced cardiomyocyte apoptosis through regulating the miR-1290/HIF3A/HIF-1α axis. The AC16 human myocardial cell line cultured under normoxic or hypoxic co… Show more

“…These cells are differentiated following transfer into mitogen-depleted media that stops their proliferation, and they express cardiac-specific proteins and maintain cardiac nuclear and mitochondrial DNA. AC16 cardiomyocytes have been used and validated in an in vitro study that examined the effect of asiatic acid of IRI [59]. The results were consistent with other studies in a rat model and an in vitro H9c2 cardiomyoblast model that also demonstrated asiatic acid attenuation of IRI [60, 61].…”

“…As for miR-1290, it was predominantly reported to act in tumor progression [ 65 , 66 ]. In addition, it has been demonstrated that the upregulated miR-1290 in non-small-cell lung cancer A549 cells could attenuate ischemia/reperfusion induced myocardial apoptosis [ 71 ]. Hence miR-1290 may be able to enhance the vascularization of ECs.…”

“…Firstly, based on the versatile activities of AA, our model may only partially explain its protective effect against hepatic I/R. Previous studies have shown that AA acted to regulate various downstream pathways such as Nrf2, AMPK, mTOR, Akt/GSK-3β/HIF-1α and miR-1290/HIF3A/HIF-1α, among many others [ 42 , 54 - 56 ]. Given that most of these pathways are critically involved in I/R, it is probable that AA may mediate hepatoprotection through these pathways besides PPARγ/NLRP3 axis.…”
