Arrestin domain‐containing protein 3 recruits the NEDD4 E3 ligase to mediate ubiquitination of the β2‐adrenergic receptor

Abstract: Prolonged stimulation of the b2-adrenergic receptor (b2AR) leads to receptor ubiquitination and downregulation. Using a genomewide RNA interference screen, we identified arrestin domaincontaining 3 (ARRDC3) as a gene required for b2AR regulation. The ARRDC3 protein interacts with ubiquitin ligase neural precursor development downregulated protein 4 (NEDD4) through two conserved PPXY motifs and recruits NEDD4 to the activated receptor. The ARRDC3 protein also interacts and co-localizes with activated b2AR. Knoc… Show more

“…To find highly conserved residues that might be important for the localization of ARRDC1 to the plasma membrane, we aligned amino acid sequences of human arrestin paralogs with ARRDC1 and with another ARRDC protein, ARRDC3, which also is known to localize to plasma membrane (Fig. 4B, Upper) (39,40). Several conserved residues (F88, G180, and N191) in the arrestin domain of ARRDC1 were identified.…”

“…It remains unclear whether WWP1 also has a role in ARMMs production. Interestingly, our previous study showed that the ARRDC1 relative ARRDC3 also interacts with members of the NEDD4 E3 ligase family (40). Through interaction with NEDD4 E3 ligases, ARRDC3 mediates ubiquitination of β2-adrenergic receptors.…”

“…In reconstitution assays, Mdm2 was shown to ubiquitinate the ␤ 2 AR, and ␤-arrestin2 was required for this event (5). Additional studies have also implicated NEDD4 as an E3 ligase for the ␤ 2 AR, and NEDD4 has been shown to interact with the receptor in a ␤-arrestin-dependent (47) and ␤-arrestin-independent manner (49). Although these varying results may be due to differences in the assay design, they may also suggest that multiple E3 ligases may be able to ubiquitinate the same receptor.…”

“…6) where the process of transporter ubiquitination begins. During the preparation of our manuscript, two studies reported the involvement of Arrdc3 in the ubiquitination and degradation (but not in internalization) of ␤2-adrenergic receptor and ␤4-integrin (55,56).…”
