Amygdalin is known as vitamain B17, and it was called laetrile. Amygdalin is composed of two molecules of glucose, one molecule of benzaldehyde which induces an analgesic action, and one molecule of hydrocyanic acid which is an anti-neoplastic compound. Amygdalin had been used to treat cancers and relieve pain. In order to evaluate whether the analgesic action of amygdalin is related with descending pain control system, we performed patch clamp study. In the present study, the modulatory effects of amygdalin on glycine-and glutamate-induced ion currents in periaqueductal gray (PAG) neurons were investigated using the nystatin-perforated patch clamp method. Continuous application of lipopolysaccharides (LPS) on PAG neurons resulted in increased glycine-induced ion current, and in decreased glutamate-induced ion current. In contrast, continuous application of amygdalin with LPS resulted in decreased glycine-induced ion current increased by LPS, and increased glutamateinduced ion current decreased by LPS in concentration-and time-dependent fashion. These results demonstrate that amygdalin modulates neuronal activity of PAG by modulation of glycine and glutamate. Based on the present results, it can be suggested that amygdalin participates in the regulation of the descending pain control system in the level of PAG neurons. The present study demonstrated that activation of the descending pain control system is one of the possible analgesic mechanisms of amygdalin.

ÖZDoğada en çok karşılaşılan siyanojenik glikozit olan amigdalin kayısı, badem, kiraz, elma, erik, armut ve şeftali gibi meyvelerin çekirdeklerinde bulunur. Amigdalin, D(-)-mandelonitril-β-D-gentiobiyozit, D(-)-amigdalin, R-amigdalin, laetril ve vitamin B 17 gibi isimlerle de anılmaktadır. Amigdalinin parçalanması enzimatik aktivite, ısıl işlem, mineral asitler ve yüksek askorbik asit konsantrasyonları ile gerçekleşebilir ve parçalanma sonucunda zehirli hidrojen siyanür oluşur. Amigdalin, alyuvar üretiminin kaybı ile karakterize edilen kansızlık, astım, yüksek tansiyon, damar sertliği, şeker hastalığı, migren ve özellikle de kanser tedavisinde kullanılmaktadır. Laboratuvar koşullarında yapılan bazı çalışmalarda tümör gelişiminin engellenmesine dair olumlu sonuçlar elde edilmiş olmasına karşın özellikle hastalığı ilerlemiş kişilerde amigdalinin tümör gelişimini önlediğine ilişkin ikna edici veriler bulunmamaktadır. Bu nedenle amigdalinin anti-kanserojen etkisinin olup olmadığı konusunun biran önce açıklığa kavuşturulması için ayrıntılı bilimsel çalışmalara ihtiyaç duyulmaktadır. Bu derlemede, amigdalinin kimyasal özellikleri, bulunduğu gıdalar, parçalanması, sağlık üzerine etkisi ve analiz yöntemlerine ilişkin bilgiler verilmiştir.Anahtar Kelimeler: Amigdalin, kimyasal özellikleri, parçalanması, sağlık üzerine etkileri AMYGDALIN AND ITS PROPERTIES ABSTRACTAmygdalin is the most common cyanogenic glycoside in nature, and it is found in fruit kernels, such as apricot, almond, cherry, apple, plum, pear and peach. Amygdalin is also called D(-)-mandelonitrile-β-D-gentiobioside, D(-)-amygdalin, R-amygdalin, laetrile and vitamin B 17 . Amygdalin can be degraded by enzymatic activity, heat treatment, mineral acids or high concentrations of ascorbic acid, and after its degradation, toxic hydrogen cyanide is formed. Amygdalin has been used to treat aplastic anaemia, asthma, hypertension, atherosclerosis, diabetes, migraine, and most important of all, cancer. Although tumour-inhibiting effects of amygdalin have been observed from some in vitro studies, there is no substantial evidence showing that amygdalin induces discrete tumour regression in cancer patients, particularly in those with late-stage disease. Therefore, further investigation is necessary to clarify whether amygdalin has an anti-carcinogenic effect as soon as possible. This paper aims to provide current knowledge about amygdalin regarding its chemical properties, presence in foods, degradation, possible health effects, and analysis methods.

Different natural products are known for their use in conventional medicine and especially for their beneficial effect on chronic diseases including autoimmune and cancer. A purified natural extract sample from the Chinese herb Tripterygium wilfordii Hook F (known as Lei Gong Teng or Thunder of God Vine) containing diterpenoids and triepoxides, was associated with significant anti-tumor effects and with encouraging results. Additionally, another natural extract from apricot kernels, Amygdalin (also known as laetrile or vitamin B17) a cyanide-containing substance, has been reported as a potential anti-cancer molecule with numerous contradictions among different published data. For many years different scientist tried to investigate in vivo, in vitro and ex vivo the efficacy of all of these molecules but they all failed in giving clear robust positive conclusions and the opinions are contradictory. Most of the studies were focusing on the apoptotic effects that can lead to the programmed cell death (PCD), a noteworthy mechanism in cancer treatment. The present review summarizes published reports and gives a better understanding of the known abilities as well as the possible role of these specific natural extract molecules in the fight of cancer treatment, cure and prevention.
