Para garantir a eficácia, a segurança e a credibilidade aos medicamentos dispensados à população, as farmácias magistrais devem aplicar as Boas Práticas de Manipulação, de acordo com a Anvisa. Ainda que limitada a aplicação integral das análises farmacopeicas nas farmácias de manipulação, a resolução estabelece os ensaios mínimos exigidos aocontrole da qualidade das preparações magistrais e oficinais. Demais testes são aplicáveis nos casos de manipulação de estoque mínimo ou análises trimestrais como forma de monitoramento do processo magistral. Neste estudo, a uniformidade de peso e de conteúdo foram avaliadas em cápsulas de ranitidina de seis diferentes farmácias magistrais em Belo Horizonte, MG, Brasil. As preparações foram Analisadas de acordo com os procedimentos de determinação de peso e de uniformidade de doses unitárias preconizados na Farmacopeia Brasileira e no Formulário Nacional. Apenas uma farmácia apresentou cápsulas com resultado insatisfatório no teste de determinação de peso. Em relação à uniformidade de doses, os produtos de todas as farmácias foram reprovados nos critérios do teste. Os resultados mostram que as cápsulas com uniformidade de peso não apresentam, necessariamente, uma distribuição homogênea do princípio ativo. O controle rigoroso do processo de manipulação é essencial para a garantia de produtos de qualidade, eficazes e seguros.

A new spectrophotometric method is proposed for the assay of ranitidine hydrochloride (RNH) in bulk drug and in its dosage forms using ceric ammonium sulphate (CAS) and two dyes, malachite (MAG) green and crystal violet (CV) as reagents. The method involves the addition of a known excess of ceric ammonium sulphate to ranitidine hydrochloride in acid medium, followed by the determination of unreacted CAS by reacting with a fixed amount of malachite green or crystal violet and measuring the absorbance at 615 or 582 nm respectively against the reagent blank. The Beer's law is obeyed in the concentration range of 0.4-8.0 μg/ ml of ranitidine hydrochloride (RNH) for RNH-MAG system and 0.2-1.6μg/ml of ranitidine hydrochloride for RNH-CV system. The molar Absorptivity, Sandell's sensitivity for each system were calculated. The method has been successfully applied to the determination of ranitidine hydrochloride in pure and dosage forms.

An environmentally friendlier, safer and saver method is described for the assay of drugs as hydrochloride salts namely metformin hydrochloride and phenylpropanolamine hydrochloride via the determination of their chloride contents. In this method, an aqueous solution of drug was treated with measured excess of silver nitrate in the presence of nitric acid, followed by the determination of unreacted silver nitrate by Volhard’s method using ammonium thiocyanate titrant and iron(III) alum indicator. To minimize the reagents consumed and wastes generated, the reactions were scaled down to less than 2 mL carried out in microcentrifuge tubes and using micropipettes for the transfer of reagents. The equivalence point was determined by spectrophotometry to diminish visual errors by reading the absorbance of red iron(III) thiocyante complex at 450 nm on microplates, which quickened the measurements of multi-samples. After validation, the method showed satisfactory accuracy and precision and was successfully applied for the assay of both drugs in raw materials, giving the results comparable to the pharmacopeial methods. In addition, the proposed assay was free from the use of harsh, toxic and environmentally harmful chemicals i.e. glacial acetic acid, acetic anhydride, mercuric acetate which are employed in the USP non-aqueous titration methods. Thus, the method is considerably safer for analysts and is a cost-effective and green analytical method suitable for a sustainable environment.
