ARF3 inhibits proliferation and promotes apoptosis in gastric cancer by regulating AKT and ERK pathway

Abstract: ADP-ribosylation factor 3 (ARF3) has confirmed participate in diverse biological processes in many cancers. However, the expression patterns and roles of ARF3 in gastric cancer (GC) remains largely unknown. In our study, by using qRT-PCR and western blot, we found that, in In GC tissues and cells, the expression of ARF3 was significantly down-regulated. Functional experiments demonstrated that ARF3 inhibited proliferation, induced cycle arrest and enhanced apoptosis of GC cells. Moreover, by performing western… Show more

“…This was mirrored in analysis of prostate cancer patients who possessed metastatic disease when the key ARF3-RAB11FIP4 cargo N-cadherin protein levels were low. In breast cancers, ARF3 upregulation was reported as associated with pro-proliferative functions 30 , while in gastric cancer, similar to our findings in prostate cells, ARF3 suppressed proliferative function and in vivo tumourigenesis 31 . Indeed, in our own analysis while ARF3 expression was commonly altered in tumour compared to normal tissue, the directionality of expression change was dependent on tumour type.…”

“…In line with our data, elongated spindles result from the depletion of the constituent Golgi proteins, which keep the integrity of the organelle in mouse oocyte 90 . The role of ARF1 in mediating Golgi morphological changes during mitosis 43 , and the function of class I ARF proteins in controlling proliferation have been reported 118 , 119 , including the ARF3 involvement in cell cycle progression and apoptosis in gastric cancer 118 . Our zebrafish data show variably penetrant effects on cell proliferation and death, especially for p.P47S and p.T32N.…”

“…Patients with bladder cancer with high levels of ARL14 methylation have a worse prognosis ( 112 ). ARF3, a homolog of ARL14, is said to express itself insufficiently in gastric cancer and may function as a possible biomarker for the prognosis of gastric cancer ( 113 ).…”
